Örökletes bőrgyógyászati megbetegedések: A genetikai hátterű bőrgyógyászati betegségek között kiemelt jelentőségű az epidermolysis bullosa (EB) csoport. EB simplexben szenvedő betegek keratin 5 és 14 génjeinek mutáció analízisét komplikáló pszeudogén kiküszöbölésére új módszert dolgoztunk ki, allél-specifikus PCR és RFLP kombinálásával. Epidemiológiai megfigyelésünk szerint hazánk egyes régióiban a csecsemőhalandóság az EB junctionalis Herlitz variánsának következtében kimagasló, ezért alapító hatás következtében domináló gyakoriságú, halmozódó mutációt valószínűsítettünk a laminin-332 heterotrimer adhéziós fehérjét meghatározó géneket érintően. EB dystrophica asszociált spinocelluláris carcinoma immunhisztokémiai vizsgálatainak eredménye felvetette, hogy a MMP (matrix metalloproteináz)-7 és MMP-13 a progresszió tumor-sejt specifikus markerei, és így a tumorterápia potenciális targetjei lehetnek. Herediter elszarusodási zavarok esetében új gének vizsgálatát vezettük be és sikeres mutációanalízist végeztünk. Immunológiai bőrgyógyászati megbetegedések: Kombinált in silico és in vitro real-time PCR analízissel bizonyítottuk a bullosus pemphigoid multikauzális kórkép ethiopathogenezisében az anello vírusok közé tartozó Torque Teno vírus szerepét.
kutatási eredmények (angolul)
Inherited dermatological diseases: The epidermolysis bullosa (EB) group is of particular importance among genetically inherited dermatological diseases. We developed a new method combining allele specific PCR and RFLP for the analysis of keratin 5 and 14 gene mutations in EB simplex patients, for the exclusion of pseudogenes. Our epidemiological survey showed that infant mortality is strikingly high in certain regions of our country due to the Herlitz variant of junctional EB, and we proposed a high frequency cumulative mutation involving laminin-332 heterotrimer adhesion protein genes through founder effect. Our immunohistochemical studies of dystrophic EB associated spinocellular carcinomas suggest that MMP (matrix metalloproteinase)-7 and MMP-13 are tumor-specific markers of progression and may be potential targets in tumor therapy. We developed new genetical assays in hereditary dyskeratoses for the mutational analysis of new genes. Immunological skin diseases: Combining in silico and in vitro real-time PCR analysis we confirmed the role of Torque Teno virus (an anellovirus) in the multicausal etiopathogenesis of bullous pemphigoid.
