Hogyan vezet az egyes idegsejtek aktivitása az agy működéséhez? Neuroanatómiai és élettani módszerekkel a kérdés a követekző elemekre bontható: Az egyes sejtek hogyan integrálják a rájuk érkező bemeneteket és állítják elő kimenetüket? Milyen kapcsolódási szabályok alapján szerveződnek a neuronhálózatok? Hogyan szabályozódik és modulálódik a neuronhálózatok működése?
Az agykéreg aktivitását magatartás-függő EEG állapotok ? információ-feldolgozó módok ? jellemzik. Ezen aktivitási állapotok közötti átkapcsolásban kéreg alatti területek, mint például a mediális szeptum és a raphe magvak, vesznek részt. Anatómiai, in vivo élettani és farmakológiai módszereket, valamint neuronhálózat modelleket tervezünk felhasználni, hogy felderítsük a hippokampális neuronok információfeldolgozásának szabályszerűségeit, illetve a mediális szeptum és a raphe magvak aktivitásának hatását. Vizsgálataink során: 1) azonosítjuk a funkcionálisan eltérő hipokampális idegsejtekre érkező bemenetek konvergenciáját; 2) tanulmányozzuk a septo-hippokampális pálya finomszerkezetét, hogy kimutassuk, a fiziológiailag heterogén szeptális sejtek egyes csoportjai mutatnak-e célelem szelektivitást; 3) a hippokampuszba vetítő szeptális sejtek élettani és farmakológiai tulajdonságait jellemezzük in vivo; 4) megvizsgáljuk a a mediális raphe szerotonerg pálya moduláló hatását hippokampális gátlósejtekre és az EEGre in vivo, helyi és szisztémás adagolt farmakonok hatására; 5) kísérleti eredményeinket felhasználva neuronhálózat modelleket készítünk a théta aktivitás kialakulásának és modulálásának modellezésére.
Summary
How do brain functions emerge from the activity of neurons? At the level of neuroanatomy and physiology the question may be spelled out as follows: How do individual neurons integrate their inputs to generate output? What are the construction rules of the networks they form? How is the activity of networks controlled and modulated?
The activity of the cerebral cortex is characterized by behaviour-dependent EEG states - information processing modes. The alteration of these states is controlled by subcortical modulatory areas, such as the medial septum and the raphe nuclei. Combining anatomical, in vivo physiological and pharmacological experiments with neuronal network modelling techniques, we will study the rules of information processing in hippocampal neurons and the modulation of their activity by medial septal and raphe afferents. We will: 1) identify the synaptic convergence and connectivity of different types of hippocampal neurons; 2) study the fine structure of hippocampal/septal interactions, revealing possible differences in the target selection of physiologically distinct medial septal neurons 3) characterize anatomical, physiological and pharmacological properties of septo-hippocampal GABAergic neurons in vivo; 4) examine the modulating effect of the median raphe serotonergic input on the activity of hippocampal inhibitory cells and the EEG in vivo, by local and systemic drug application; 5) using our data from the experiments, neuronal network models will be constructed to study network mechanisms of the generation of the theta rhythm.

Kimutattuk, hogy a szeptum és a hippokampusz között a HS sejtek szintjén reciprok gátló kapcsolat áll fent. A HS sejtek érzékelik a hippokampális aktivitás szintjét, elsősorban szeptális gátlás alatt állnak és a szeptumon kívül a hippokampusz távoli területeire is vetítenek ezáltal a szeptum és az egész hippokampusz működését összehangolják. A theta aktivitással együtt előforduló gamma aktivitás kialakításában elsősorban a PV tartalmú kosársejtek játszanak szerepet, aktivitásukat, és így a gamma aktivitást opiát receptorok útján szabályozni lehet. A szeptum és a hippokampusz kölcsönhatását vizsgálva kimutattuk, hogy a medialis septum HCN ioncsatornát es/vagy parvalbumin fehérjét kifejező gátlósejtjei vezérlik a hippocampalis theta ritmust. Részletes CA1-es hálózati modell segítségével jóslatokat fogalmaztunk meg a théta és magasabb frekvenciájú oszcillációk lokális mechanizmusaival, kölcsönhatásaival és extrahippokampális modulációjával kapcsolatban. A felszálló szerotoninerg rendszerben kimutattunk egy az eddig ismerteknél gyorsabb, glutamátot használó hatékony moduláció-típus létezését.

We demonstrated that the reciprocal interaction between the medial septum and the hippocampus is realized at the cellular level via the HS cells. They detect the level of hippocampal activity, are under primarily septal inhibitory control and besides the septum they project to remote hippocampal areas, synchronizing the activity of this two regions. The theta concurrnet gamma activity is primarily controlled by the PV containing basket cells. These cells and thus gamma activity can be modulated via opiate receptors. Studying the interaction of t emedial septum and the hippocampus we proven that HCN/PV expressing medial septal inhibitory neurons drive the hippocampal theta rhythm. Simulations of a detailed network model of area CA1 resulted in predictions regarding the local mechanisms, interactions, and extrahippocampal modulation of theta and higher frequency oscillations. We demonstrated a quick, glutamatergic component in the ascending modulatory raphe-hippocapal serotonergic projection.
