A fertőző megbetegedések paleopatológiai rekonstrukciója az utóbbi évtizedek kiemelt jelentőségű kutatási irányvonala volt mind a magyarországi, mind a nemzetközi paleopatológiai kutatásban. A kutatások anyaga a váci Fehérek templomának kriptájából feltárt 265 természetesen mumifikálódott emberi maradvány. A korábbi vizsgálatok igazolták a tervezett antropológiai, biometriai, patológiai, laparoszkópiás, radiológiai, valamint archaikus DNS vizsgálatok kivitelezhetőségét.
Célunk, hogy egy ősi, a gyógyszerek és a modern orvosi működés hatásától mentes populáció esetében végezzünk vizsgálatokat több betegségre hajlamosító polimorfizmus, ill. néhány krónikus betegségben szenvedőkben halmozottan előforduló patogén tekintetében. A multidiszciplináris pályázat egyedülálló abban a tekintetben, hogy egy viszonylag rövid időszak népességét reprezentálja meglehetősen nagy egyedszámmal (n=265). A kutatás magába foglalja a népesség paleoepidemiológiai érintettségét a kiválasztott virális és bakteriális fertőzöttség tekintetében, a fertőzöttségre való hajlamosságot csökkentő enzimpolimorfizmusok vizsgálatát és a populáció általános populációgenetikai elemzését.
Az alábbi kérdéskörök molekuláris genetikai vizsgálatát tervezzük:
-A mannóz-kötő fehérje polimorfizmus szintjének vizsgálata archaikus és modern mintákban.
-CCR5 HIV coreceptor gén polimorfizmus vizsgálata.
-A Kaposi sarcomában etiológiai szerepet játszó humán herpeszvírus 8 specifikus DNS kimutatása.
-A szifilisz nyomainak kimutatása T. pallidum specifikus molekuláris eljárással.
-A humán mitokondriális DNS hipervariábilis régióinak szekvenciaszintű meghatározása és az egyedek mitokondriális haplotípusainak meghatározása a múmiák fog illetve csontszövetéből nyert DNS extraktumban.
-Az Y kromoszóma markereinek vizsgálata a múmiák fog illetve csontszövetéből nyert DNS extraktumban.
Lényeges, hogy a modern orvosi ellátás miatti szelekció nem érvényesült a vizsgálandó csoportnál, így a betegségek (ma már nem vizsgálható) természetes lefolyást mutatták, teret engedve a genetikai és környezeti tényezők közötti „valódi” kölcsönhatásnak. Munkánk emiatt az orvos-biológiai kutatásokban nóvum.
A résztvevő kutatók publikációiban is tükröződő gyakorlati tapasztalat biztosítja, hogy a tervezett kutatás korrekt kivitelezésre alkalmas. Az infrastrukturális háttér biztosított, a pályázott összeg részben a vegyszerekre és reagensekre fordítandó.

One of the key topics of the worlds’ leading palaeopathological researches has been the reconstruction of past epidemics. We propose a multidisciplinary study of 256 naturally mummified bodies came to light in the Dominican Church of Vác, Hungary. Previous studies have shown the usability of laparoscopic, X-ray, CT and aDNA studies on them.
With the study of a population lived before the era of extensive use of antibiotics and modern medicines, we aim to learn about polymorphism related to disease susceptibilities and some pathogens frequently occur in chronically ill patients. Our project is unique in focusing on a fairly large population restricted to a rather short period, incorporating its degree of infection by the selected microorganisms, detection of enzyme polymorphisms related to disease susceptibilities, and population genetic studies.
With isolation of the archaic mummy sample’s ancient DNA content, topics include:
- Level of mannose binding lectin (MBL) polymorphism,
- Level of CCR5 HIV coreceptor gene polymorphism,
- Isolation of HHV8 specific DNA, which has a role in Kaposi Sarcoma,
- Isolation of T.pallidum specific DNA, the causative agent in syphilis,
- Sequencing of HVR I- II of human mtDNA and haplotyping human mtDNA,
- Study of human Y-chromosomal STR markers.
As the selective effect of modern medical treatments did not affected this population, unlike today the natural progress of diseases can be postulated, which makes possible to study the “real” interaction of genetic and environmental factors. That means a novelty in medical biological studies.
The rather large number of contributors and their extensive experiences reflected in their publications gives a proof of the implementation of the designed experimental work, so does the availability of the infrastructural background.

A kutatásokat a váci Fehérek templomának kriptájából feltárt természetesen mumifikálódott emberi maradványon végeztük. A kutatás anyaga egyedülálló lehetőséget biztosított abban a tekintetben, hogy egy viszonylag rövid időszak népességét reprezentálja, meglehetősen nagy egyedszámmal (n=265). Fontos kiemelni, hogy a modern orvosi ellátás miatti szelekció nem érvényesült a vizsgált népességcsoportnál. A kutatás során egy ősi, a gyógyszerek és a modern orvosi működés hatásától mentes populáció esetében végeztünk vizsgálatokat több, betegségre hajlamosító polimorfizmus, ill. néhány krónikus betegségben szenvedőkben halmozottan előforduló patogén tekintetében. Ugyancsak célunk volt a népességcsoport multidiszciplináris, mikro-történeti, biológiai-patológiai rekonstrukciója, egészségügyi helyzetének feltérképezése.
