Az idegrendszerben az endo és exokannabinoidok fő sejtszintű hatása a neurotranszmitter felszabadulás gátlása, melyet preszinaptikusan fejtenek ki. Korábbi vizsgálatainkban kimutattuk, hogy a hippokampuszban a kannabinoidok gátolják a GABA és a glutamát felszabadulást idegi aktivitás során a hippokampuszban és a striatumban, és míg a GABA felszabadulásra gyakorolt hatást a CB1 kannabinoid receptorok közvetítik, a glutamát felszabadulásra gyakorolt hatás független a CB1 receptoroktól, és azt egy, ma még ismeretlen, kannabinoid-szenzitív receptoriális mechanizmus közvetíti. Más kutatócsoportok vizsgálatai is kimutattak CB1 és CB2 receptor-független kannabinoidhatásokat az agyban és a periférián, ezek közös nevezőre hozása azonban nehéz, mivel a különböző kutatócsoportok a kannabinoid ligandok eltérő skáláját használták. Ezért fő célkitűzésünk, hogy egy, az eddigieknél lényegesebben komplett farmakológiai analízissel és CB1 receptor génkiütött egerek felhasználásával összevessük a klasszikus CB1 receptort és a feltételezett CB3 receptor(ok)at, és ezáltal farmakológiai bizonyítékot szerezzünk az új receptor létéről/nemlétéről, majd ennek ismeretében azonosítsunk új, preszinaptikus kannabinoid receptorhatásokat az agyban. Ezek közé tartozik a hippokampális szerotoninfelszabadulást esetlegesen befolyásoló preszinaptikus kannabinoidreceptor, valamint a kannabinoidok GABA, glutamát és dopaminfelszabadulásra gyakorolt hatása a nucleus accumbens-ben. Emellett a kannabinoidok és az abuzusdrogok (nikotin, kokain, amfetamin, MDMA, ethanol, opiátok) neurokémiai egymásrahatását is tanulmányozni szeretnénk a nucleus accumbensben, mélyebb betekintést nyerve ezáltal a kannabinoidok addiktív tulajdonságaiba.
Summary
The main cellular action of exo- and endocannabinoids in the brain is the inhibition of neurotransmitter release with a presynaptic site of action. In previous studies we showed that cannabinoids inhibit GABA and glutamate release in the hippocampus and striatum, respectively, and whereas their inhibitory effect on GABA release is mediated by CB1 cannabinoid receptors, that of glutamate release is independent from CB1 receptor activation and is conveyed by an unknown cannabinoid sensitive receptorial mechanism. Other studies also revealed CB1 and CB2 receptor independent actions of cannabinoids in the brain and in the periphery, however it is difficult to reconcile them, since different working groups used different selection of ligands. Thus the main objective of the study is to perform a comprehensive pharmacological analysis with a wide range of ligands and the use of CB1 receptor deficient mice lines to compare the classical CB1-receptor, and the putative CB3 receptor(s) and thereby to obtain a pharmacological proof on the existence /absence of non-CB1-cannabinoid receptors. Using this information for receptor identification, the next aim is to reveal, new presynaptic cannabinoid actions in the brain; i.e. presynaptic cannabinoid receptors involved in the regulation of serotonin release in the hippocampus as well as GABA, glutamate and dopamine release in the nucleus accumbens. In addition, we also plan to investigate the neurochemical interaction of abuse drugs (nicotine, cocaine, amphetamine, MDMA, ethanol, opiates) in the nucleus accumbens in order to get a deeper insight to the addictive properties of cannabinoids.

Kimutattuk, hogy bár a kannabinoidok GABA felszabadulásra gyakorolt hatását a hippokampuszban a CB1 receptorok közvetítik, ezek a hatások részben fennmaradnak a CB1 receptor genetikai törlése esetén is, valószínűleg egy „tartalék” kannabinoid receptor feldúsulása révén. Elsőként írtuk le és jellemeztük a kannabinoidok gátló hatását a szerotonin felszabadulásra a hippokampuszban. A kannabinoidok hatását a CB1-receptorok közvetítik, és az a szerotonerg terminálisoknak elsősorban egy szubpopulációjára terjed ki. Kimutattuk, hogy az endokannabinoidok a hippokampuszban a bazális IL-1beta produkció szabályozásában is szerepet játszanak, mégpedig stimuláló jelleggel és a P2X7 receptorok közvetítésével. Eredményeink elsőként igazolják, hogy a nucleus accumbens drog addikcióban kiemelten fontos szerepet játszó dopaminerg végződéseiből a kannabinoidok nemcsak a dopaminerg neuronok ventralis tegmentum-ban elhelyezkedő sejttestjeinek stimulálásával, hanem a nucleus accumbensen belüli hatással, dizinhibíciós mechanizmussal is képesek dopamint felszabadítani. Leírtuk a noradrenalin és acetilkolin felszabadulás frekvenciafüggő kannabinerg modulációját a prefrontális kéregben. Kimutattuk, hogy GPR3 receptor genetikai törlése az agyi monoamin tartalmak csökkenéséhez és ezzel korreláló magatartásváltozásokkal jár a szorongás és a depresszió állatkísérletes modelljeiben.

We showed that the effect of cannabinoids on GABA release in the hippocampus is mediated by CB1-cannabinoid receptors. However, these effects are partly maintained after genetic deletion of CB1 receptors, and probably due to a residual, „backup” cannabinoid receptor, which is overexpressed in CB1 knockouts. We reported for the first time the inhibitory effect of cannabinoids on serotonin release from the hippocampus. The action of cannabinoids is mediated by CB1 receptors, but affects only one subpopulation of serotonergic nerve terminals. We showed that endocannabinoids stimulate basal IL-1beta production in the hippocampus, partly with the participation of P2X7 receptors. We provided the first neurochemical evidence that the activation of CB1 cannabinoid receptors leads to the augmentation of [3H]dopamine efflux via a local GABAA receptor-mediated disinhibitory mechanism in the rat nucleus accumbens. In addition, the frequency dependent modulation of noradrenaline and acetylcholine release by cannabinoids is characterized in the prefrontal cortex. We also showed that genetical deletion of GPR3 receptor leads to the depletion of monoamine content in the brain and consistent alterations of behavior in animal models of anxiety and depression.
