Az enzimműködés megértése az elmúlt évszázadban folyamatosan fejlődött, de távol áll még a tökéletestől. Jóval a három dimenziós szerkezetmeghatározó eszközök, (NMR. Röntgen krisztallográfia) elterjedése előtt az enzimműködés funkciónális vonatkozásait a fehérje flexibilitás alapján képzelték el. Jelenleg nagy mennyiségű szerkezeti információ érhető el, és a szerkezetek flexibilis természete is bizonyítást nyert. Megközelítésünk szerint az enzimfunkció felderítése és megértése a szerkezeti stabilitás és flexibilitás finom egyensúlyán alapul. Komplex módszerek együttesével tervezzük ezt az összefüggést felderíteni különböző szerveződési szinteken: a domének szubmolekuláris szintjén, az amionsavak atomi szintjén, és a kvantumeffektuson szubatomi szintjén.
A szerkezeti elemek az enzimműködésben játszott szerepének felderítésében a következő célokat tűztük ki.
1) A modularitás szerepének felderítése a multidomének enzimek specificitásában, szabályozásában és stabilitásában
2) Az enzimreakciók rendellenes hőmérsékletfüggésének térszerkezeti hátterének vizsgálata
3) A kvantumhatások, mint az alagúteffektus szerepének tisztázása az enzimatiku skatalízisben (H-, H+ és e- transzfer reakciók esetén)
A célok eléréséért modellenzimekkel végzünk vizsgálatokat, amelyeken már tapasztalokat szereztünk, és a következő eredményeket tervezzük elérni:
1) A komplement rendszer működésének és szabályozásának megértése molekuláris szinten , és a reakciómechanizmus alaposabb megismerése egy multidomén szerin proteáz enzimcsalád esetén (C1r, C1s, MASP-1,2,3)
2) A konformációs stabilitás, flexibilitás és enziműködés közötti összefüggések általános felderítése modellenzimek felhasználásával
3) Az anomális hőmérsékletfüggés kvantifikálása enzimatikus H-, H+ és e- transzfer reakciók esetén, valamint az alagúteffektus enzimreakciók lejátszódásában való hozzájárulásának ellenőrzése.

The understanding of enzymatic action has been steadily developing throughout the last century, however, it is far from complete. Long before the spread of three dimensional structure determining tools such as NMR and X-ray crystallography , the explanation of functional aspects of enzymatic actions has been derived using the concepts based on protein flexibility. At present, vast amount of structural information is available and the flexible nature of structures has also been evidenced. Our approach of exploring and understanding enzyme function is based on the assumption of a fine balance of structural stability and flexibility. We plan to apply an arsenal of complex methods in order to investigate this relationship at different structural levels: the sub-molecular level of domains, the atomic level of amino acids and the subatomic level of quantum effects.
To elucidate the role of structural elements in enzymatic functions we set our goals:
1) Explore the role of modularity in the specificity, regulation and stability of multidomain enzymes.
2) Examine the structural background of the anomalous temperature dependence of enzymatic reactions.
3) Investigate the role of quantum effects such as tunnelling in enzymatic catalysis (in H-, H+ and e- transfer reactions)
To achieve these goals we plan to work with model enzymes we have accumulated expertise with and are thriving to reach the following results:
1) Understand the function and regulation of the complement system at molecular level, and obtain an insight into the mechanism action in the case of a family of multidomain serine proteases as C1r, C1s, MASP-1,2,3
2) Draw general conclusions on the relationship among conformational stability, flexibility and enzyme function using model enzymes.
3) Quantitate the anomalous temperature dependence of enzymatic H-, H+ and e- transfer reactions and identify the degree of the contribution of quantum effects e.g. tunnelling.

Az elmúlt 3 év koherens kutató munkája során születtek speciális tudományos eredmények és levontunk ezekből általános következtetéseket is. Munkánk mérlege a nemzetközi folyóiratokban megjelent 30 közlemény összesen 130 IF-al. Molekuláris immunológiai kutatásaink keretében meghatároztuk 4 komplement proteáz és a C1-inhibitor szerkezetét, különösen az utóbbi hozott számunkra nagy nemzetközi elismerést. A szerkezetek és funkcionális eredményeink alapján általánosan elfogadott aktiválási modellt dolgoztunk ki a komplement rendszer lektin útjának szabályozási mechanizmusára. Jelentősnek tartjuk a C1-inhibitor heparin által történő potencirozásának mechanizmusára javasolt, szerkezeti alapú modell kidolgozását, a flagellin fehérje egyik rendezetlen szakaszának export szignálként történő azonosítását (szabadalom is született belőle), a foszfoglicerátkináz enzim domén záródásban résztvevő allosztérikus jeltovábbító hálózat azonosítását, az enzimaktivitás rendhagyó hőmérsékletfüggésének a konformációs flexibilitás alapján történő értelmezését a izopropilmalát dehidrogenáz esetében, átmeneti zóna felfedezését a rendezett és rendezetlen szerkezetet kódoló aminósav szekvencák között. A komplement fehérjék és funkcionális komplexeik, a flagelláris rendszerek, multidomén enzimek együttes vizsgálata lehetővé tette a fehérjék önszerveződésével, a molekuláris szintű felismeréssel és az allosztérikus jeltovábbítás mechanizmusával kapcsolatos általános következtetések levonását.

We have determined the structure of C1-Inhibitor and four complement proteinases: C1r, MASP1, MASP2 in zymogen form and MASP2 in activated form. Based on our structural and functional studies we concluded a mechanistic model for the activation of the lectin pathway of the complement system. We also devised a structure based model for the heparin potentiation of C1-Inhibitor. An intrinsically disordered sequence of the bacterial flagellin protein was identified as an export signal (patented). Other significant achievements: the mapping of an allosteric network involved in the ligand induced hinge closure of phosphoglycerate kinase, the interpretation of the odd temperature dependence in the catalytic activity of isopropylmalate dehydrogenase in terms of concerted conformational fluctuations, discovery of the twilight zone between amino acid sequences encoding ordered and disordered conformations. Our coherent studies on the functional protein complexes of the complement system, on flagellar systems, multidomain enzymes enabled us to make some general conclusions regarding the self assembly, recognition and allosteric behaviour of proteins and protein complexes.
