Jelen pályázatom a pozítívan értékelt EURY pályázatom (EYA 049968) egy új változata, melyben a tervezett munka egy részét kihagytam a költségvetés csökkentése érdekében.

A sejt számára komoly kihívást jelent ha a replikációs apparátus DNS-hibához érve megakad. Élesztőben a Rad6-Rad18 ubiquitin-konjugáló komplex a károsított DNS replikációjának három alternativ útját irányítja: a Rad5-függő hibamentes, a polimeráz éta-függő hibamentes, és a polimeráz zéta-függő mutációt okozó átírást. A károsodott DNS mutációt okozó átírása karcinogenezist indukál, míg hibamentes replikációja hozzájárul a genetikai stabilitás megőrzéséhez.

Kutatási eredményeim nagymértékben hozzájárultak a károsított DNS replikációja terén az elmúlt négy évben bekövetkezett áttöréshez, amely elvezetett a polimeráz éta és zéta funkciójának megértéséhez és feltárta, hogy a polimeráz éta mutációja miként vezet a Xeroderma pigmentosum nevű bőrrák betegség kialakulásához.
Nem ismert azonban, hogy az élesztő Rad5-függő út miként írja át hibamentesen a károsított DNS-t, illetve, hogy hasonló folyamat működik-e emberekben. Jelen pályázatom célja az, hogy ezekre a fontos kérdésekre választ adjon.

Elsőként a Rad5 fehérje kölcsönható partnereit tervezem azonosítani és jellemezni, majd a Rad5-függő DNS replikációt in vitro homogenitásig tisztított fehérjékkel rekonstruálni. Megvizsgálom, hogy a human HLTF tumor szupresszor gén, amely az élesztő Rad5 homologja, a polimeráz étához hasonlóan részt vesz-e a károsodott DNS hibamentes replikációjában csökkentve ezáltal a mutagenesist és karcinogenezist..

Tervezett kísérleteimtől azt remélem, hogy fényt derítenek azokra a DNS károsodást toleráló folyamatokra, amelyek csökkentik a rákos folyamatok kialakulásának valószínűségét.
Summary
The current proposal is a new version of my highly scored EURY grant proposal (EYA 049968), in which I propose to carry out only part of the original work to reduce the cost of the proposed research.

The stalling of DNA replication machinery that occurs as a consequence of encountering unrepaired DNA damage is a challenging problem for cells. In yeasts, the Rad6-Rad18 ubiquitin-conjugating complex governs three alternative pathways of replication of damaged DNA: the Rad5-dependent error-free, the DNA polymerase eta dependent error-free, and the polymerase zeta dependent error-prone damage bypass. In humans, increased error-prone bypass of DNA lesions causes increased mutagenesis, and as a consequence, a rise in the incidence of cancers, whereas error-free replication of damaged DNA contributes to genetic stability.

My research work has contributed to the great progress, which has been made in the past four years toward the understanding of the function of polymerase eta and zeta and of unravelling how mutations in polymerase eta cause Xeroderma pigmentosum, a cancer prone syndrome.

However, it is still not known how the Rad5-dependent pathway stimulates error free replication of damaged DNA in yeasts, and whether a similar pathway in humans operates. The goal of my proposed project is to answer these important questions.

First, I propose to identify new protein elements in the yeast Rad5-pathway and to reconstitute it in vitro by highly purified protein factors. Second, I plan to examine human HLTF, a recently identified tumour suppressor, which shares high degree of sequence homology with yeast Rad5. I will test the possibility that HLTF, as the human homologue of yeast Rad5, similarly to human Poleta, may prevent carcinogenesis by promoting error-free replication of damaged DNA.

My study will shed more light on the molecular events of DNA damage tolerance pathways, which prevent mutagenesis and carcinogenesis.

A kijavítatlan DNS hibák gyakran gátolják a DNS replikációt. Saccharomyces cerevisiae-ben a károsított DNS replikációja történhet a hibák direkt átírására specializálódott DNS polimerázokkal illetve az ún. Rad5-függő posztreplikációs mechanizmus segítségével, amely az újonnan szintetizálódott szál a DNS hibával szemben kialakult diszkontinuitásait szünteti meg. Míg a polimerázok közreműködésével történő hibaátírás részletesen jellemzett ismereteink a posztreplikációs mechanizmusról igen korlátozottak. Projektünk célja az volt, hogy betekintést nyerjünk az élesztő Rad5-függő hibaátírásba. Kutatásaink során felfedeztük, hogy a Rad5 fehérjének egy speciális DNS helikáz aktivitása van, amely a replikációs villa visszafordítására specializálódott. Modell replikációs villa struktúrákon a Rad5 képes széttekerni mindkét minta/leány szálat majd hibridizálni egymáshoz a leány és a mintaszálakat. Ebből arra következtethetünk, hogy a Rad5 a DNS mintaszál-váltást katalizálva segíti elő a károsodott bázis hibamentes átírását. A továbbiakban sikerült azonosítanunk az élesztő Rad5 fehérjének két emberi homológját, a HLTF és SHPRH fehérjéket, is. A HLTF gyakran inaktiválódik vastagbél és gyomor rákban, míg az SHPRH a melanomák mintegy tíz százalékában mutálódik, amely tumor szuppresszor funkciójukra utalhat. Eredményeink szerint a HLTF és az SHPRH a károsított DNS hibamentes átírásában játszik kulcsszerepet, amely megmagyarázhatja tumor szuppresszor funkciójukat.
Results in English
Lesions in the template DNA strand block the progression of the replication fork. In yeast, replication through DNA lesions is mediated by different Rad6-Rad18-dependent means, which include translesion synthesis and a Rad5-dependent postreplicational repair pathway that repairs the discontinuities that form in the DNA synthesized from damaged templates. Although translesion synthesis is well characterized, little is known about the Rad5-dependent pathway. The aim of our project has been to give insight into the yeast Rad5-dependent postreplicational repair pathway. We found that yeast Rad5 has a DNA helicase activity that is specialized for replication fork regression. On model replication fork structures, Rad5 concertedly unwinds and anneals the nascent and the parental strands without exposing extended single-stranded regions. These observations provide insight into the mechanism of postreplicational repair in which Rad5 action promotes template switching for error-free damage bypass. Also, we have identified and characterized two functional human homologues of yeast Rad5, the human HLTF and SHPRH proteins. HLTF is frequently inactivated in colorectal and gastric cancers while SHPRH is mutated in about ten percent of melanoma cell lines which has indicated a potential tumor suppressor function for these proteins. Our finding that HLTF and SHPRH play a role in error-free postreplication repair of damaged DNA is in keeping with its cancer-suppression role.
