A tudatos idegrendszeri működés egyik alapja a specifikus és nem specifikus, talamuszból eredő információk agykérgi integrációja. A specifikus, vagy elsőrendű talamikus relémagvak működése intenzíven kutatott terület. A nem specifikus, vagy magasabbrendű talamikus magvak aktivitását szabályozó mechanizmusok alapjai azonban alig ismertek, noha ezek sérülése komoly kognitív zavarokkal jár. Az eredeti OTKA pályázat célja e magasabb rendű magvak működésének strukturális és funkcionális alapjainak megértése.
Az OTKA project során új kollaboráció alakult a Laval egyetemen dolgozó Martin Deschenes munkacsoportjával a magasabb rendű talamikus magvak funkciójának felderítéséhez kapcsolódó specifikus kérdések megválaszolására. Ennek során az egyik legtöbbet kutatott szenzoros rendszer (a patkány bajusz érzékelő rendszere) és in vivo intracelluláris elvezetések segítségével a csak a magasabbrendű talamikus magvakra jellemző fő serkentő és gátló pályák funkcióját a szenzoros stimuláció adta kontrollált körülmények között vizsgáljuk. A résztvevő kutatócsoportok egymást kiegészítő tudományos tapasztalatai lehetővé teszi a talamusz sejt és hálózat szintű tulajdonságainak korrelált anatómiai és fiziológiai kutatását. Eredményeink megváltoztathatják a talamikus funkcióról, illetve a talamusz és agykéreg hierarchikus viszonyáról kialakult képünket, valamint hozzájárulhatnak a patológiás körülmények között keletkező abnormális fiziológiai aktivitás alapjainak megértéséhez.
Summary
Conscious cognitive functions depend on the cortical integration of ascending information carried by specific and non-specific thalamocortical afferents. The specific or first order thalamic relay nuclei are well studied but the principles governing the activity of non-specific or higher order nuclei are relatively unexplored. The primary goal of the original OTKA project is to reveal the structural and functional basis of the rules of operation in higher order thalamocortical circuits by studying its major excitatory and inhibitory afferents.
During the OTKA project a new collaboration was formed with Martin Deschenes (Universitée Laval, Quebec) to answer specific questions of higher order thalamic functions. Using the best characterized sensory system, the vibrissal system of the rat, and in vivo intracellular recordings the function of the major excitatory and inhibitory pathways in the thalamus can be studied in a replicable and controllable way. Complementary expertise of the participating teams allows using correlated anatomical and physiological methods to study the cellular and system properties of thalamic networks. The results may challenge our current view of thalamic functions, the hierarchical relationship of thalamus and cortex, and will enhance our understandings of the generation and spread of abnormal physiological activity in pathological conditions.

Az OTKA pályázat során leírtunk és karakterizáltunk egy új gátlás-típust a talamuszban. E gátlórendszer axonvégződései és az általuk közvetített gátlás különbözött a talamuszban jól ismert gátlórendszerek tulajdonságaitól. Az axonterminálisok ultrastruktúrája és az általuk beidegzett célelemek hatékony gátlásra utaltak. Élettani kísérletek igazolták az anatómiai predikciókat és kimutatták, hogy ez a gátlás típus hatékony információ átvitelre képes magas frekvenciás impulzusok esetén is, mikor az ismert gátlópálya hatékonysága jelentősen csökken. Az új gátlás-típus képes megakadályozni a beidegzett talamikus sejtek működését, illetve képes kiváltani a karaterisztikus visszacsapó választ. Az új gátlás-típust sikerült azonosítani több pálya esetén, köztük főemlősökben a Parkinson-kórban érintett talamikus bemenetek esetében is. A pályázat során részletesen vizsgáltuk az új gátló pálya eredő sejtjeinek szerkezetét és működését. Egy új gátlópálya leírása, melyet egyedi működési mechanizmusok jellemeznek felveti a lehetőségét e pálya szelektív modulációjának. Olyan drogok, melyek ezen a speciális gátlóterminálison hatnak segíthetnek azon tünetek enyhítésén, melyeket e pályák aberráns aktivitása okoz (pl. Parkinson-kór, krónikus fájdalom).
Results in English
During the OTKA project we discovered and characterized a novel inhibitory element in the thalamus. The structure of the nerve endings, their mode of action and the activity of the nerve cells were all different from the previously described inhibitory pathways in this brain centre. The anatomy of the nerve terminals and the nerve elements they contacted indicated powerful inhibitory action. Physiological measurements verified the anatomical predictions and demonstrated that these connections faithfully transfer inhibitory signals even at very high frequency, when the effectiveness of other inhibitory pathways is much reduced. We demonstrated that these inhibitory inputs are indeed able to silence thalamic neurons or induce strong, so-called, “postinhibitory rebound” activity. The morhology and the actvity of the parent cells of these pathways has also been extensively characterized. We described several of these pathways in different thalamic nuclei most importantly in those known to be involved in Parkinson’s disease. Discovering a separate class of inhibitory pathways in the thalamus with distinct mode of action raises the hope for their selective modulation. Drugs which acts on these terminals can help to alleviate the symptoms linked to the aberrant activity of these specialized inhibitory pathways.
