HYPOPHYSIS ADENILÁT-CIKLÁZ AKTIVÁLÓ POLIPEPTID (PACAP) HATÁSÁNAK VIZSGÁLATA KÜLÖNBÖZŐ TÉNYEZŐK KÖVETKEZTÉBEN KIALAKULÓ COCHLEARIS- ÉS RETINALIS KÁROSODÁSBAN

A PACAP-ot 1989-ben izolálták birka hypothalamusból, tagja a szekretin/glukagon/ VIP peptidcsaládnak. Felfedezése óta számos kísérletben bebizonyították már neurotrofikus és neuroprotektív hatását mind in vitro, mind in vivo kísérletekben. Munkacsoportunk is évek óta foglalkozik a PACAP neuroprotektív hatásának vizsgálatával. Kimutattuk, hogy védő hatást fejt ki fokális cerebralis ischaemiában, Parkinson-kór, Huntington-chorea, neurotrauma modellben és retina károsodásban, valamint in vitro és in vivo kísérletekben is bizonyítottuk antiapoptotikus hatását. Továbbiakban PACAP hatásának vizsgálatát retinalis- és cochlearis károsodásokban kívánjuk folytatni. Vizsgálataink egyik részében ciszplatin- és gentamicin-indukálta halláskárosodásban vizsgáljuk a PACAP védő hatását, mivel ezen szerek mellékhatásai nagy problémát jelentenek a klinikai gyakorlatban. Vizsgálni kívánjuk az állatok hallását agytörzsi válaszok detektálásával, valamint otoakusztikus emisszió mérésével, és ezeket a méréseket szövettani és molekuláris biológiai vizsgálatokkal is kiegészítjük. A kísérleteink másik részében az újszölöttkori toxikus és hypoxiás cochlearis- és retinakárosodást vizsgáljuk, mivel ezen elváltozások is nagy problémát jelentenek a klinikumban. Korábban már kimutattuk, hogy a PACAP képes kivédeni a glutamát-indukálta retinalis degenerációt, de a hatásmechanizmust pontosan még nem ismerjük, ezért céljaink között szerepel ezen kísérletek folytatása, dózishatásgörbe vizsgálata, valamint a morfológiai vizsgálatok mellett molekuláris biológiai módszerek alkalmazása. A retinadegeneráció mellett vizsgáljuk a glutamát ototoxikus hatását is. Végül kísérleteinket az újszülöttkori hypoxia következtében kialakuló érzékszervi károsodások vizsgálatával tennénk teljessé.
Kísérleteink során szeretnénk adatokat nyerni a PACAP hatásáról különböző eredetű hallás és látáskárosodást modellező állatkísérletekben. A PACAP hatásmechanizmusa pontosan még nem ismert, ezért a funkciónális vizsgálatok mellett morfológiai és molekuláris biológiai módszerek eredményeit is közölni kívánjuk, hogy elősegítsük az anyag mielőbbi klinikai alkalmazását.

Examination of PACAP neuroprotective effect in cochlear and retinal degeneration

PACAP was first isolated from ovine hypothalami in 1989, it is a member of the secretin/glucacon/VIP family. Numerous studies show that PACAP is a neuroprotective peptide both in vitro and in vivo experimental conditions. Recently, we have shown that PACAP has neuroprotective effect in a rat model of focal cerebral ischemia, Parkinson’s and Huntington’s disease, diffuse axonal injury and retinal degeneration, and we have also shown that it has antiapoptotic effect in in vitro and in vivo experimental conditions. The aim of the present study is to investigate the effect of PACAP in a rat model of retinal and cochlear degeneration. We examine the effects of PACAP in cisplatin- and gentamicin-induced ototoxicity, because these represent major problems in the clinical practice. We investigate the auditory brainstem responses (ABR) and the otoacoustic emission (OAE) of the animals, and we complete this with morphological and molecular biological experiments. In the other part of the study we examine the retinal and cochlear degeneration of newborn rats induced by monosodium glutamate-treatment and hypoxia. Recently, we have shown that PACAP attenuates glutamate-induced retinal degeneration, but the exact pathomechanism is not known. Therefore, the aim of the study is to continue these experiments with molecular biological examinations. Besides the retinal degeneration we examine the ototoxic effect of the glutamate treatment. Finally, we finish our experiments with the examination of the effects of neonatal hypoxia in the retinal and cochlear degeneration.
We would like to show the effects of PACAP in retinal and cochlear degeneration induced by different toxic agents. We would like to investigate the pathomechanism of PACAP in these experimental conditions in order to get more detailed insight of the neuroprotective effects exerted by PACAP.

A kutatási támogatás segítségével 48 publikációt közöltünk (IF:124,798). A pályázatban megfogalmazott két legfontosabb cél a PACAP cochleáris és retinális károsodásokban kifejtett hatásainak vizsgálata volt, amihez legszorosabban 21 dolgozat kapcsolható (IF: 62,474). Cochleáris vizsgálataink során a PACAP megvédte a belső fül sejtjeit oxidatív stressz-indukálta apoptózissal szemben. A vad és PACAP KO egerek cochleájának összehasonlítása során kimutattuk, hogy a vad egerek szignifikánsan több PAC1 receptort és kevesebb Ca2+-kötő fehérjét expresszálnak, bizonyítván azt, hogy a PACAP-nak fontos szerepe van a szőrsejtek toxikus hatásokkal szembeni védekezésében. Igazoltuk a PACAP retinoprotektív hatását MSG-indukálta és ischemia-indukálta károsodással szemben mind in vivo, mind in vitro kísérletekben. Leírtuk továbbá az újszülöttkori MSG-kezelés, a perinatális asphyxia, az anyai megvonás és az ingergazdag környezet hatásait is a retina fejlődésére. Bár a vad és PACAP KO egerek retinájában morfológiai eltérés nem látható, a PACAP hiányos egerek szignifikánsan nagyobb károsodást szenvedtek ischemiát követően, mint vad társaik. Vizsgáltuk továbbá a PACAP jelenlétét különböző emberi szövetekből és testváladékokból. Bizonyítottuk a PACAP protektív hatását különböző vese és bélkárosodásban, valamint általános citoprotektív hatását különböző sejtkultúrákban, többek között human retinális pigment epithel sejtekben, mely hozzájárulhat a PACAP retinoprotektív hatásának megismeréséhez.

We published 48 publications with the help of this grant (IF: 124,798). 21 (IF: 62,474) publications are closely related to the main focus of this grant, namely, the effects of PACAP on cochlear and retinal lesions. We showed that PACAP protects cochlear cells against oxidative stress-induced apoptosis. We compared the inner ear of wild type and PACAP-deficient animals. PACAP-deficient animals showed significantly decreased PAC1 receptor expression. In contrast, hair cells of PACAP-deficient mice showed more pronounced Ca2+-binding protein expression compared with wild-type mice providing further evidence for the important role of PACAP in hearing processes. We proved the retinoprotective effect of PACAP against MSG- and ischemia-induced degeneration in vivo and in vitro. We also described the effect of MSG-treatment, perinatal asphyxia, maternal separation and enriched environment on retinal development of newborn rats. We did not find significant morphological difference in the retina between the wild type and PACAP-deficient animals, but PACAP-deficient mice suffered a significantly greater damage after ischemia compared with wild animals. In addition, we investigated the presence of PACAP in different tissue samples and body fluids. Our experiments also showed the protective effect of PACAP in renal and intestinal damage and in different cell cultures, including human retinal pigment epithelial cells, which can be an additional factor in the retinoprotective effects of PACAP.
