A neuroepithelium adhézióját és a szinapszisok összekapcsolódását az idegrendszerben elsősorban a cadherin-catenin és a nectin-afadin komplexek által kialakított adhéziós kapcsolatok biztosítják. A cadherinek közül elsődleges szerepe van az N-cadherinnek (Cdh2), aminek szerepét a neuritkinövésben, a szinapszisok kialakulásában és érésében, valamint a hippocampális LTP kialakításásban korábban in vitro kísérletek bizonyították. Egérben azonban a gén kiütése korai lethalitást okoz (E10) a szív fejlődési rendellenességei miatt, így a molekula neurális fejlődésben betöltött szerepéről keveset tudunk. Célom, hogy az egeret, mint modellállatot alkalmazva áttekintő képet kapjunk az N-cadherin expressziójáról és funkciójáról a teljes egéragy prenatális embrionális fejlődése során és a posztnatális fejlődés első hónapjában a hippocampusban, Az expressziót RNS és fehérje szinten is vizsgáljuk in situ hibridizációs és immunhisztokémiai festésekkel valamint a kettő kombinációjával. A funkciót transzgénikus egerek előállításával vizsgáljuk. A fehérjét tetraciklinnel indukálható módon és szövetspecifikusan termeltetjük túl GAD65 (GABAerg sejtek), CamkIIa (glutamáterg sejtek), illetve nestin promoterrel (korai neuroepithelium) vezérelt palzmidkonstrukciók injektálásával. A transzgénikus állatokat ezután komplex molekuláris, sejt- és szövetszintű morphológiai elemzésnek (és szükség esetén magatartási tesztetknek) vetjük alá. Az általunk előállított vektorrendszer a későbbiekben jelentősen egyszerűsíti más molekulák funkcionális analízisét.

Cadherin-catenin and nectin-afadin based adherens junctions are the major force in cell adhesion and synapse formation in the CNS, with N-cadherin (Cdh2) being one of the main cadherin components. Knocking out N-cadherin causes lethality at E10 in the mice due to failure of heart differentiation, hence the protein’s role in later neural development remains largely unknown. In vitro experiments however, have shown its importance in neurite outgrowth, synapse formation and maturation, and in hippocampal LTP. My goal is, to investigate the expression and function of N-cadherin during mammalian neural development using a transgenic mouse model. Expression will be examined at RNA and protein level using whole-mount and floating vibratome section in situ hybridization, immuncitochemistry and the combination of these two. For the functional studies, I will create transgenic mice lines that overexpress N-cadherin or its dominant negative version in a Tet-inducible, tissue-specific manner. Tissue specificity will be provided by the CamkIIa (glutamatergic cells), GAD65 GABAergic cells) and nestin (early neuroepithelium) promoters. The transgenic animals will be analysed by a combination of anatomical, histological and molecular methods with the primary target being prenatal development and postnatal neuronal differentiation in the hippocampus. With a simple cut and paste replacement cloning the created driver and expression vectors will be very useful in functional studies of other molecules.

A kitűzött cél az N-cadherinnnek a gátló, GABAerg internuronjainak fejlődésében betöltött szerepének vizsgálata. Ezt expressziós analízissel és egy Tet-indukálható kettős transzgénikus modell megteremtésével kívántuk vizsgálni. Az expressziós analízist be is fejeztük, sőt 12 II-es típusú klasszikus cadherinre is kiterjesztettük, ami szintén majdnem teljesen kész. A Tet-indukálható rendszerhez beszerzett kereskedelmi forgalomban kapható bicisztronikus plazmid azonban nem produkált N-cadherin és/vagy eGFPexpressziót az elkészült transzgénikus egérben. A projekt közben megjelent közleményben ezért a heterokromatikus részbe történő beépülést és az emiatti géncsendesedést tarják felelősnek. Ennek kiderülése után az utolsó évben rohamtempóban meghonosítottuk Magyarországon elsőként, az in utero elektroporációt, amivel tranziensen és mozaikosan túlexpresszáltatjuk az N-cadherint a gangliondomb sejtjeiben. A funkcióvesztés vizsgálatára megszereztük a 2009-ben elérhetővé váló kondicionális N-cadherin-/- mutáns egeret és ezt a Gad65-GFP egértörzsünkkel kereszteztük a született GABAerg sejtek könnyű nyomonkövetése céljából, majd a Kanadából megkapott Dlx-Cre vonalhoz kereszteztük és a tripla transzgénikus egerek most nőnek. Heteken belül megkezdődik a 3. generáció visszakeresztezése a homozigóta Ncad-/- egerekkel és a GABAerg sejtek migrációjának és differenciációjának analízise az időközben megklónozott sejttípus-specifikus markerekkel.

The main goal of the project is to reveal the potential function of N-cadherin in the development of GABAergic interneurons using expressional analysis and by creating a tetracycline-inducible double transgenic mouse model. The expression analysis was carried out successfully. Moreover, we expanded it to 12 members of the type II classic cadherin family also presumed to play an important role in neuronal development. The planned ectopic N-cadherin or reporter eGFP expression was however missing in the doxycycline-induced double transgenic animals. A paper which appeared during the course of the project revealed that the preference of the bicistronic vector to incorporate into heterochromatic regions is responsible for this silencing phenomenon. This forced us to find alternative approaches. In the last year of the project, we established for the first time in Hungary the in utero electroporation technique for overexpressing locally the N-cadherin in the ganglionic eminences where GABAergic interneurons are born. To obtain loss-of-function data we obtained the floxed N-cadherin-/- conditional KO mice which became commercially available in 2009. We crossed them to our Gad65-eGFP line for easy tracking of the GABAegic differentiation. Finally we crossed these mice to Dlx-Cre to inactivate N-cadherin specifically in the ganglionic eminences. Results coming from the analysis of GABAergic interneuron differentiation in these triple transgenic mice will happen within weeks.
