Az akut szervi, szöveti ischaemia-reperfusio (I/R) nem minden aspektusában tisztázott, vizsgálata korunk keringési betegségeinek ijesztő mértékű elterjedtsége miatt is indokolt. Korábban a végtagi I/R pathomechanizmusát tanulmányoztuk. A vér áramlási tulajdonságát (haemorheologia) leíró faktorok fontos információt nyújtanak e folyamatokban, változásaikat több fizikai, morfológiai, funkcionális, metabolikus és hormonális tényező befolyásolja, emellett fajspecifikus és méréstechnikai különbségeket mutatnak.
Sok adat mutatja, hogy a rheologiai paraméterekben nemi különbségek vannak, melyek meghatározóak lehetnek a klinikai kép és a terápiás hatékonyság vonatkozásában. Ez fölveti a kérdést, hogy a gonádok csökkent, vagy hiányzó működése milyen mértékű hatással bírhat az egyes haemorheologiai paraméterekre, illetve egy “másodlagosan” bekövetkező végtagi I/R milyen további hatásokat rejthet magában. Ezekkel összefüggő és kevéssé vizsgált kérdés a gonádok I/R-s károsodása.

Célkitűzéseink:
1. Haemorheologiai méréstechnikák laboratóriumi állatfajokra való adaptációs munkáinak folytatása, finomítása.
2. Haemorheologiai összehasonlító mérések végzése (filtrometer, cell transit analyzer, ektacytometria)
3. Az ivarmirigyek I/R-s károsodásának morfológiai, haemorheologiai vizsgálata (inbred egér, patkány).
4. Csökkent, vagy hiányzó gonádműködés végtagi I/R-s folyamatokra kifejtett hatásának haemorheologiai vizsgálata (outbred patkány, inbred beagle kutya).
Summary
Beside the alarming large incidence of circulatory diseases in our days, acute ischemia, ischemia-reperfusion (I/R) of organs and tissues and their pathophysiological processes have not been completely clarified yet. Earlier we have investigated the limb I/R.
Parameters describing rheological properties of blood give valuable information on these processes, they are determined by several physical, morphological, physiological, metabolic and hormonal factors, and they express species specific and methodological alterations as well.
Hemorheological factors also have differences in gender that influence the clinical state and the therapeutical effectiveness. Question raises, whether lost or altered gonad function has an effect on the individual hemorheological parameters, if yes in what degree, and are there further consequences during a ‘secondary’ limb ischemia-reperfusion. Furthermore, a purely investigated question is the I/R injury of gonads.

The objectives of this research:
1. Continuation and refinement of our adaptation work on hemorheological measurement techniques on laboratory animal species.
2. Comparative methodological measurements of erythrocyte deformability (filtrometry, cell transit analyzer, ektacytometry)
3. Morphological and hemorheological examinations of I/R in gonads (inbred mice, rats).
4. Effects of lost or altered gonad function on limb I/R and its hemorheological examinations (outbred rats, inbred beagle dogs).

A kutatási program fő célkitűzései: I. Haemorheologiai összehasonlító vizsgálatok, II. A gonadectomia (GoE), valamint a III. GoE-t követően előidézett végtagi ischaemia-reperfusio (I/R) és a IV. Gonad I/R haemorheologiai vonatkozásai voltak. Ad I.: Új adatokat nyújtottunk laboratórium állatfajok (CD outbred patkány, inbred beagle kutya) haematocrit és vérviszkozitás kapcsolatához, az "optimális haematocrit" meghatározáshoz a vörösvérsejt (vvs) deformabilitás és vvs aggregatio faji különbségei mellett. A vérmintavételi hely, az anticoagulans, a mintatárolás és mintakezelés vonatkozásában újabb adatokat szolgáltattunk az állatkísérletes haemorheologiai filtrometriás és az ektacytometriás mérések optimalizálásához. A vizsgált laboratóriumi állatfajok haemorheologiai paramétereinek jelentős, a fajok között nem konzekvensen változó nemi különbségeit mutattuk ki. Ad II.: Kimutattuk, hogy laboratóriumi patkányokon a GoE-t követő 2. és 3. postoperativ (p.o.) hónapban jelentős vvs aggregatio és deformabilitás változás jelentkezett. Ad III.: Patkányban az 1 órás hátsó végtagi I/R hatására az 1-3. p.o. napokon mutatkozott, jelentős vvs deformabilitás romlás, nőstényeknél nagyobb mértékben. A nőstények GoE+I/R csoportjában a postischaemiás értékek mutatták a legjelentősebb romlást. Ad IV.: Patkányokon előidézett 30 perces testis ischaemia után a vvs deformabilitás az egy órás reperfusio végén szignifikáns romlást mutatott, a vvs aggregatio fokozódását az 1. p.o. napon találtuk.
Results in English
The main objects of the research were: (I) Comparative hemorheological measurements; the hemorheological effects of (II) gonadectomy (GoE); of (III) hind limb ischemia-reperfusion (I/R) performed after GoE; and of (IV) gonadal I/R. Ad I: We could provide new data on the relation of hematocrit to blood viscosity and ’optimal hematocrit’ of laboratory animals (CD outbred rat, inbred beagle dog) besides describing inter-species differences of red blood cell (RBC) deformability and RBC aggregation. Concerning the blood sampling site, the anticoagulant usage, the sample handling and storage we could provide novel data for standardization of hemorheological measurements and for optimalization of filtrometry and ektacytrometry techniques. Ad II: In the 2nd and 3rd postoperative month after GoE marked changes of RBC deformability and aggregation were demonstrated in rats. Ad III: The 1-hour hind limb ischemia and the following reperfusion resulted in significant impairing of RBC deformability on the 1st to 3rd postoperative days in rats, which changes were more expressed in female animals. In female GoE+I/R group the postischemic result showed the worst results. Ad IV: In rat model of 30-minute testicular ischemia RBC deformability impaired in a highly significant manner by the 60th minute of the reperfusion, and increasing of RBC aggregation index was observed by the 1st postoperative day.
