A gombatermesztésben jelentős károkat okozó ún. „zöldpenész” tünetegyüttes kialakulásáért a Trichoderma nemzetségbe tartozó fonalasgombák tehetők felelőssé. Az elmúlt években a hazai gombatermesztők a zöldpenészes megbetegedés megjelenését és terjedését észlelték csiperkén és laskán, ami agresszív Trichoderma törzsek hazai megjelenésére utal.

Jelen pályázatban célul tűztük ki hazai csiperkekomposztból és a laska termesztésére alkalmazott szubsztrátból származó Trichoderma törzsek taxonómiai, mikopatológiai és epidemiológiai vizsgálatát. Részletesen tanulmányozni fogjuk az izolált törzsek biodiverzitását, genetikai variabilitását, azonosítjuk a csiperke és laska zöldpenészes megbetegedését hazánkban kiváltó agresszív Trichoderma fajokat és felmérjük a termesztési körülményekre történt specializáltságuk mértékét. Vizsgáljuk a komposzt- és szubsztrátmintákból származó izolátumok fungicidekkel szembeni érzékenységét. Tanulmányozzuk a zöldpenész kialakulásának potenciális virulenciafaktorait, in vitro antagonizmus-vizsgálatokat hajtunk végre a termesztett gombák és vad típusú, valamint bizonyos enzimek termelésében deficiens, illetve túltermelő mutáns Trichoderma törzsek között az egyes enzimek fertőzési folyamatban betöltött szerepének tisztázása céljából. A tervezett epidemiológiai felmérés az agresszív törzsek nyomonkövetését, a zöldpenész fertőzés lehetséges forrásainak azonosítását célozza.

A kutatómunka várható eredményei elmélyítik a Trichoderma nemzetséggel kapcsolatos tudományos ismereteket, ezen túl hasznos információkkal szolgálnak hatékony megelőzési stratégiák és kémiai, ill. integrált védekezési eljárások kidolgozásához.
Summary
Filamentous fungi belonging to the genus Trichoderma are responsible for the so called „green mold” disease causing significant losses in mushroom production. During the last years, Hungarian producers noticed the occurrence of green mold both for the white button mushroom and the oyster mushroom, suggesting the emergence of aggressive Trichoderma strains in Hungary.

The proposed project aims the taxonomical, mycopathological and epidemiological investigation of Trichoderma strains isolated from Hungarian Agaricus compost and Pleurotus substrate samples. The biodiversity and genetic variability of the isolated strains will be studied in details, the aggressive Trichoderma species responsible for the green mold diseases of Agaricus and Pleurotus in Hungary will be identified and the level of their substrate specialization will be determined. The susceptibility of the isolates deriving from compost and substrate samples will be examined towards fungicides. The potential virulence factors related to green mold will be studied. In vitro antagonism experiments will be carried out between the grown mushrooms and the wild-type Trichoderma strains as well as mutants with increased or decreased production of distinct extracellular enzymes in order to clarify the role of certain enzymes played in the infection process. The proposed epidemiological investigation aims the follow-up of aggressive strains and identification of the possible sources of infection.

The expected results of this project will contribute to the scientific knowledge about the genus Trichoderma, furthermore, it will provide useful information for the development of efficient preventive measures as well as chemical and integrated control strategies.

Eredményeink alapján a termesztett csiperke zöldpenészes fertőzését Magyarországon a Trichoderma aggressivum f. europaeum okozza, míg a laskagomba zöldpenészes megbetegedéséért a T. pleurotum és T. pleuroticola fajok felelősek. Környezeti tényezők micéliumnövekedésre gyakorolt hatásának vizsgálata alapján a laskagombatermesztésben az átszövetési szakasz utáni fázisra 15-18°C közötti hőmérséklet és 8-9 közötti pH javasolható, mely lassíthatja a zöldpenészt okozó Trichoderma törzsek növekedését. Kidolgoztunk egy antagonizmus-tesztek képanalízisén alapuló módszert a zöldpenészt okozó Trichoderma törzsek agresszivitási fokának számszerűsítésére. Enzimológiai vizsgálataink alapján a két laskagombapatogén Trichoderma faj számos ponton eltérő stratégiát alkalmaz a termesztésben fennálló körülményekhez történő alkalmazkodás céljára. A laskagombapatogén Trichoderma fajok epidemiológiai nyomonkövetésére kidolgoztunk egy multiplex PCR-alapú technikát, mely lehetővé teszi a két faj gyors kimutatását és egymástól történő elkülönítését. A módszer segítségével igazoltuk, hogy a laskagomba zöldpenészes fertőzésének terjesztésében szerepet játszhatnak a termesztő telepeken jelenlevő rovarok, és azonosítottuk a fertőzés egy lehetséges forrását: a T. pleuroticola faj előfordul a vadon termő laskagomba természetes környezetében is. Eredményeink alapján bizonyos, gombatermesztésből izolált baktériumok (pl. Bacillus törzsek) ígéretesek lehetnek a zöldpenésszel szembeni biológiai védekezés céljára.
Results in English
Our results revealed that the green mould infection of cultivated champignons in Hungary is caused by Trichoderma aggressivum f. europaeum, while T. pleurotum and T. pleuroticola are responsible for the green mould disease of oyster mushroom. Based on the effects of environmental factors on mycelial growth, a temperature between 15-18°C and a pH between 8-9 can be suggested for the period of spawn run in order to slow down the growth of Trichoderma strains. We established a method based on the image analysis of antagonism tests for the quantification of the aggressivity level of Trichoderma strains causing green mould disease. Our enzymological studies revealed that the two oyster mushroom pathogenic Trichoderma species apply different strategies for the adaptation to the cultivation environment. A multiplex PCR-based technique has been developed for the epidemiological monitoring of oyster mushroom pathogenic Trichoderma species, which enables the fast detection of the two species and their differentiation from one another. This method was applied to prove that the insects present in mushroom farms can play a role in the dissemination of the disease and to identify a potential source of the infection: T. pleuroticola was found to occur also in the the natural environment of wild-growing oyster mushroom. Based on our results, certain bacteria isolated from mushroom cultivation (e.g. Bacillus strains) are promising candidates for the biological control of green mould disease.
