Jelen pályázatunk célja a nagyobb méretű testidegen részecskékkel szemben immunsejtek közreműködésével kialakuló, tokképző reakció molekuláris alapjainak tisztázása Drosophila modellszervezeten. Ez a védekező reakció, amely Drosophilában specializálódott vérsejtek, a plazmatociták és a lamellociták közreműködésével valósul meg, hasonlít a gerinces szervezetekben a makrofágok és a T sejtek részvételével zajló granulómaképzéshez, ezért a Drosophila kiváló modellszervezet a veleszületett immunitás ezen folyamatának a vizsgálatában. Drosphilában eddig nem sikerült lárvális vérsejt alpopulációkra jellemző markermolekulákat azonosítani, ami megnehezíti az immunsejtek differenciálódási vonalainak, valamint a differenciálódás folyamatának és az effektor vérsejtek funkcióinak a vizsgálatát. A kutatócsoportunk által azonosított vérsejteken megnyilvánuló markermolekulák közül több lamellocita-specifikusnak bizonyult. Ez az eszköz egyedi lehetőséget biztosít számunkra a lamellociták, valamint a lamellociták közreműködésével lezajló tokképző reakció vizsgálatához. A lamellocita antigének előzetes vizsgálata azt mutatja, hogy meghatározó szerepük lehet a sejtek differenciálódásának szabályozásában és a tokképző reakcióban. A tokképző reakció folyamatának jobb megértése érdekében ezért tovább jellemezzük ezeket a molekulákat.
A támogatás időtartama alatt a következő célokat szeretnénk elérni:
- Az eddigi eredmények alapján a fő lamellocitaspecifikus antigennek tekinthető L1 (Atilla) molekula
további jellemzése biokémiai, klasszikus genetikai és komplex genomikai vizsgálatokkal.
- A lamellocita differenciálódáshoz és a tokképző reakcióhoz vezető fő jelátviteli utak azonosítása.
- Két további, általunk azonosított lamellocita antigénnek, az L4 és az L6 molekuláknak a tokképző
reakcióbajn betöltött szerepének vizsgálata.

The long term goal of this proposal is to decipher the molecular mechanisms that underline the immunity-related cellular reaction to large particles, the encapsulation reaction, using Drosophila as a model system. This protective response, with the contribution of specialized blood cells in Drosophila, the plasmatocytes and the lamellocytes, has some similarities to granuloma formation by mammalian macrophages and T-cells therefore Drosophila is a good model organism to study this path of the innate immune response. So far very few blood cell markers have been identified in Drosophila, which has hampered the study of their lineage, differentiation and functions. Recently, we have identified hemocyte markers some of them expressed in lamellocytes. This tool gave us a unique chance for studies on encapsulation reaction, with special emphasis on the role of lamellocytes. Preliminary definition of the identified lamellocyte markers showed that they could be involved in the regulation of lamellocyte differentiation and in the regulation of encapsulation reaction. We decided therefore to characterize these molecules further with the aim to get better unsderstandig of the encapsulation reaction.
Our aims for the funding period are:
- Definition of the function of the main lamellocyte antigen identified so far , the L1 (Atilla), using a combination of biochemical, classical genetic and a complex genomic approach.
- To identify the signaling pathways leading to differentiation of lamellocytes and to encapsulation.
- To study the involvement of two newly identified lamellocyte markers, L4 and L6, in the regulation of encapsulation.

A Drosophila melanogaster paraziták elleni tokképző reakcióját vizsgáltuk az immunsejteken általunk korábban definiált molekulák felhasználásával. Azonosítottuk a paraziták petéit tokba záró lamellocitákon kifejeződő fehérjét kódoló atilla gént. Az Atilla fehérje egy glikozilfoszfatidilinozitol horgonyozó hellyel rendelkező transzmembrán molekula, a Ly6 szupercsalád tagja, az első olyan lipid raftokkal asszociált fehérje, mely Drosophila vérsejtjein fejeződik ki. A Drosophila melanogaster genomjában 24 atilla-szerű gént találtunk. Az atilla gén közelében azonosítottuk a minos inszerciót, mely az atilla gén lamellocita specifikus enhanszerét csapdázva egyedi eszközként szolgál a lamellociták in vivo nyomonkövetésében. A lamellocitákon kifejeződő L4 antigénről megállapítottuk, hogy az a myospheroid gén által kódolt βPS integrin. Genetikai sejtvonal jelöléssel azt találtuk, hogy a molekulát kifejező sejtek a tokképző reakció során morfológiai változáson mennek át, lamellocitákká alakulásuk közben elveszítik fagocitáló képességüket, alátámasztva a veleszületett immunitás sejtes elemeinek nagyfokú morfológiai és funkcionális plaszticitását. Az L2 molekula a lamellocita sejtvonal irányban véglegesen elkötelezett sejtekben feltehetőleg az aktinnal képez molekula-komplexet. A Drosophila lárva szesszilis vérsejtjeiről megállapítottuk, hogy funkcionálisan egységes szövetet képeznek, melyből származó sejtek szolgálnak a parazita darázs petéje elleni immunválasz elsődleges forrásául.

Our studies focus on the regulation of cell mediated immunity in Drosophila model organism by the aid of the molecular markers -, in particular the lamellocyte specific L1, L4 and L2 molecules - defined by us previously. The atilla gene - encoding for the L1 protein - is expressed by lamellocytes and their precursors, cells, forming multilayer capsules around parasite eggs. The atilla gene product is a glycosylphosphatidylinositol anchored cell-surface protein, a member of the Ly6 superfamily, the first molecule that has been identified as a cell surface molecule associated with lipid rafts in Drosophila blood cells. We found 24 atilla-like genes in the genome, organized in four clusters. A minos insertion discovered in the neighbourhood of the atilla gene is suitable for in vivo detection of lamellocytes. The L4 protein is an integrin βPS encoded by the Drosophila myospheroid gene. Our genetic lineage tracing experiments showed that hemocytes expressing L4 undergo marked morphological and functional changes, the observation, that revealed the morphological and functional plasticity of cellular components of the innate immune system in Drosophila. The L2 molecule most likely forms a molecular-complex with actin at the terminal stage of lamellocyte differentiation. These molecular markers also helped us to define the sessile hemocytes as a functional hematopoietic compartment serving as the main source of lamellocytes in the course of the cell mediated immune response.
