Csoportunk fő érdeklődési területe a növényi sejtciklus szabályozásának molekuláris háttere, az ilyen irányú kutatások több mint tizenöt éves múltra tekintenek vissza. PhD hallgatóként a különböző lucerna CDK-k kölcsönható partnereinek élesztő kettős-hibrid rendszer felhasználásával történő azonosítása lett a feladatom. Doktori dolgozatomban valamint a 2006 áprilisában a The Plant Journalben megjelent cikkemben azokról a munkákról számoltam be, amelyek a lucerna CDK inhibitor (KRPMt) részletes jellemzését jelentették. A téma legfontosabb újdonsága a KRPMt foszforilációs szabályozása. Sikerrel mutattunk ki egy aktiváló foszforilációt, azonban továbbra is rejtve maradt mind az MSCPK3 (kalcium-függő fehérje kináz), mind a CDK-k foszforilációjának pontos helye és különböző hatása, annak ellenére, hogy bizonyítottnak tekinthetjük, hogy mind az A-típusú mind a B-típusú CDK-k képesek megfoszforilálni az inhibitor fehérjét. Az eddigi munka logikus folytatásaként a következő célkitűzéseket fogalmazhatjuk meg:

1. Melyik aminosav a MsCPK3 foszforiláció helye?
2. Különbözik-e ettől az A-típusú illetve B-típusú CDK foszforiláció?
3. Mik a hatásai ezen poszttranszlációs módosításoknak?
4. Szerepet játszik-e a 26S proteoszóma a KRPMt lebontásában?
5. A megszerzett ismeretek alapján elkészített foszforilálhatatlan, illetve foszforilációt mimikáló mutánsok, mutatnak-e aktivitásbeli változást in vitro kináz reakciókban?
6. Okoznak-e ezek a mutánsok fenotípus változást transzgenikus növényekben?

We have long been fascinated by the intricacy of cell division. During the last two decades have the building blocks of a molecular system and the machinery that regulates the cell cycle been revealed. Recent studies have shown that CDK inhibitors can also have a tremendous impact on cell cycle progression in plants and thus on plant growth and development.
In plants, cell cycle progression can respond to hormonal and stress factors such as auxins, abscisic acid and osmotic shock which are all known to induce Ca2+-dependent signalling pathways. The role of the Ca2+-status in the division of plant cells has been studied in a limited number of experimental systems. Among the Ca2+-binding sensory proteins, the calmodulin like domain protein kinases (CPKs) play a role in the transmission of stress and hormonal signals. In my thesis and in my article in Plant Journal I reported the cloning of an alfalfa CDK inhibitor (KRPMt) cDNA that encodes an active inhibitor protein with a differential effect on various kinase complexes. Phosphorylation of the KRPMt protein by an alfalfa MsCPK3 recombinant kinase results in the increase of KRP function in vitro. This finding supports a hypothesis that modulation of CDK activities by inhibitor protein as a substrate of the Ca2+-dependent kinase provides a molecular mechanism for crosstalk between Ca2+-signalling and cell cycle control.
This findings and the fact that CDKs are also able to phosporylate KRPMt arose the following questions:
1. On wich amino acid residue of KRPMt is phosphorylated by MsCPK3?
2. Are the phosphorylation sites of Medicago A- and B-type CDKs different from this/these?
3. What are the functional consequences of these posttransltional modifications?
4. Is the 26S proteosome involved in the degradation of KRPMt protein?
5. How behave the dephospho-mutant and phospho-mimicry mutant KRPs in kinase assays in vitro?
6. Have these mutants phenotype in transgenic alfalfa and tobacco plants?

Megállapítottuk, hogy az MsCPK3 a KRPMt 91. szerin aminosavát foszforilálja. Kísérleteink során bebizonyosodott, hogy az A-típusú CDK-k ugyanezen az aminosavon foszforilálják a lucerna CDK inhibitor fehérjét. A B-típusú CDK foszforiláció különbözőséget vagy azonosságát technikai problémák nem tették lehetővé, lévén ezen kinázokkal nem tudtunk megfelelően erős kináz aktivitást detektálni a KRPMt foszforilációja esetén. Többszöri kísérletek ellenére sem sikerült ubiqitinálást kimutatni sem a vad típusú sem a foszforilációt mimikáló rekombináns fehérjék esetében. Elkészítettük a 91. szerin aminosav foszforilációt mimikáló glutamát valamint aszpartát mutánsait. Valamint a nem foszforilálható alanin mutánst is. Ezen rekombináns fehérjék egyike sem volt foszforilálható sem az MsCPK rekombináns fehérjével, sem az A-típusú CDK-kal. Kináz gátlási kísérleteink során a foszforilációt mimikáló szerin aszpartát csere jelentös mértékben (kétszeresére) fokozta az inhibitor fehérje gátló képességét. Elkészítettük a mutáns fehérjék növényi expressziójához szükséges vektorokat, melyekkel dohány levél korongokat transzformáltunk. A GUS pozitív kalluszok regenerációja folyamatban van, azonban sajnálatos módon a legígéretesebb, legerősebb expressziót mutató kalluszok esetében jelentős degenerációt tapasztaltunk nem képesek gyökeret létrehozni. A kutatás idő előtt lezárásra kerül, mert külföldre távoztam és a halasztási kérelmemet nem támogatta az OTKA Bizottság Elnöke.

We found that the MsCPK3 Ca-dependent protein kinase could phosphorylate the KRPMt CDK inhibitor on a serine residue at position 91. Our experiments demonstrated that the A-type CDKs phosphorylate the same amino acid residue in alfalfa CDK inhibitor proteins. We were not able to reveal the kinase activity of the B-type CDK on KRPMt due to the fact that we could not purify sufficiently strong activity for clarifying KRPMt phosphorylation event. We made 91 serine phosphorylation mimicking mutants: 91Ser-91Glu and 91Ser-91Asp, and phosphoinactive 91Ser-91Ala mutants as well. None of those recombinant proteins were phosphorylated by MsCPK3 or A-type CDKs. We could not detect ubiqitination and the subsequent degradation of either the wild-type or the phosphorylation mimicking and phosphoinactive recombinant proteins. In kinase inhibition experiments, the 91Ser-91Asp recombinant protein showed significantly (twice) increased inhibitory effect on A-type CDKs. We have created the plant expression vectors with the wild type and mutant KRPMts, which were used for tobacco leaf discs transformation. The regeneration of whole plants from GUS positive calli is in progress, but unfortunately, the most promising calli -showing the strongest expression of GUS - have significant degeneration: they can not form roots. The time before the research is completed, I left for abroad and the application for postponing the project was not supported by the president of the OTKA committee.
