A “Miért kezdődik el az embriógenezis” munkatervben olyan gének szerepét tervezzük megérteni, amelyek funkciójára szükség van az embriógenezis elkezdődéséhez. A géneket a muslica (Drosophila melanogaster) olyan domináns negatív típusú nőstény-steril (Fs) mutációival (KetelD, KavarD és HorkaD) azonosítottuk, amelyek bár lehetővé teszik küllemre ép peték képződését, és megtermékenyülését, ám a petékben nem kezdődik el az embriógenezis, jelezve, hogy az Fs mutációkkal azonosított ép géneknek szerepük van az embriógenezis elkezdődésében. Az Fs mutációkból kiindulva megklónoztuk az ép géneket. Célunk molekuláris sejtbiológiai szerepük megismerése. A KetelD mutációk az importin-β-t kódoló gént azonosítják. Mi mutattuk meg, hogy importin-β nem csak a sejtmagi fehérje importhoz szükséges, hanem a sejtmaghártya összeálláshoz is. Szeretnénk megérteni, hogy miként szabályozott a Ketel gén expressziója, és hogy mely gének funkciója és miként okoz halált importin-β hiányában. A KavarD mutációk az anyai eredetű α-tubulin4 képződését kódoló αTub67C gént azonosítják. Megmutattuk, hogy az α-tubulin4 az interpoláris mikrotubulusok gyors képződéséhez szükséges, és ahhoz, hogy kapcsolódjanak a sejtmaghártyához. Tisztázni reméljük, hogy mitől különleges α-tubulin4. HorkaD a transzkripciós terminációs faktor 2 (TTF2) képződését kódoló TTF2 gént azonosítja. Kérdés, miként vezet a HorkaD kódolt mutáns TTF2, valamint a TTF2 hiánya kromoszóma instabilitáshoz, a nőstény-sterilitás újfajta típusához.
Summary
In the “What makes embryogenesis commence?” project we plan to study the mechanism of the commencement of embryogenesis. We have identified genes with dominant negative female-sterile (Fs) mutations in Drosophila melanogaster: KetelD, KavarD and HorkaD. The Fs mutations allow the production and fertilization of seemingly normal eggs, however embryogenesis does not commence inside showing that the Fs-identified normal genes play role in commencement of embryogenesis. We have already cloned the above genes and plan to understand their molecular functions. The KetelD Fs mutations identify the importin-β coding gene. We showed that importin-β is not only a key component of nuclear protein import but is also essential for reassembly of the nuclear envelope at the end of mitosis. We are set up to understand how expression of the Ketel gene is achieved and through what mechanism does the lack of importin-β lead to death of the zygotes. The KavarD mutations identify the so-called maternal α-tubulin4 isotype coding αTub67C gene. We have shown that α-tubulin4 is required for rapid formation of the interpolar microtubules and also for contact between the nuclear envelope. We plan to find out what makes α-tubulin4 special. HorkaD identifies the transcription termination factor 2 (TTF2) coding TTF2 gene. The question we hope to answer is this: how does the HorkaD encoded mutant TTF2 and also the lack of TTF2 lead to chromosome instability and the origin a novel type of female-sterility.

A “Miért kezdődik el az embriógenezis” pályázat keretében a Drosophila három génjének szerepét értettük meg. A géneket domináns negatív típusú nőstény-steril mutációkkal azonosítottuk. Belőlük kiindulva klónoztuk meg ép géneket, ismertük meg szerepüket. Importin-b. A KetelD mutációk az importin-b-t (imp-b) kódoló Ketel gént azonosítják. Leírtuk, hogy imp-b nem csak a sejtmagi fehérje import, hanem a sejtmaghártya-, valamint a magorsó-képződés szereplője. - Meghatároztuk: miként szabályozott a Ketel gén expressziója, szövet-specifikus kifejeződése. - Leírtuk: az imp-b a leghosszabb élettartamú fehérje a sejtekben. - Megmutattuk: a Ketel génnek szerepe van a mitokondrium-biogenezisben. - Az imp-b-GFP-vel „kivilágított” sejtmagpórus-komplexek mozgása alapján a sejtmag pozicionálás új módját írtuk le. a4-tubulin. A KavarD mutációk az anyai eredetű a4-tubulin képződését kódoló aTub67C gént azonosítják. A KavarD-k alapján kiderítettük, hogy - a mikrotubulusok (MT-ok) felszínén az a-tubulinok Glu426-jai a kinezinek kötődési helyei. - Új módszert (konfokális reflexiós mikroszkópia) írtunk le a MT-ok mentén történő transzport vizsgálatára. Lodestar. A HorkaD mutáció a helikáz-szerű géncsalád egyikét azonosítja. Funkciója ahhoz kell, hogy - a cohesin komplexek a kromoszómákhoz kapcsolódjanak. - Leírtuk, hogy a HorkaD-kódolt A777T-Lodestar mutáns molekulák ragadósak, rendellenes kromoszóma szegregációt okoznak.

The “Why does embryogenesis commence” project focused on three Drosophila genes. They were identified by dominant negative female-sterile (Fs) mutations. The Fs mutations were starting points for molecular analyses of gene functions. Importin-b. The KetelD Fs mutations identify the importin-b (imp-b) encoding Ketel gene. Imp-b is a component of not only nuclear protein import but also nuclear envelope and spindle formation. We - determined the mode of Ketel gene expression in the DNA and in tissue levels, - showed that imp-b is the longest lived protein in the cells, - reported that it is engaged in mitochondria biogenesis. - Starting from the motion of imp-b-GFP highlighted nuclear pore complexes we described a novel nuclear positioning mechanism. a4-tubulin. The KavarD Fs mutations identify the aTub67C gene that encodes a4-tubulin. Analysis of the KavarD mutations revealed that - the Glu426 side chains of the a4-tubulins are the positions on the surface of the microtubules (MTs) where kinesins bind to the MTs. - We described a novel method (confocal reflection microscopy) to study transport along the MTs. Lodestar. The HorkaD Fs mutation identifies the lodestar gene, a member of the helicase-related family. Its function is required - to place the cohesin complexes onto the chromosomes. - The HorkaD-encoded A777T-Lodestar mutant molecules are “sticky” and bring about abnormal chromosome segregation.
