A hazánkban honos Centaurea sadleriana föld feletti részének főzetét a Dél-Alföldön tradicionálisan felhasználják a juhok sebeinek gyógyítására. A növény kémiai összetételét, hatását korábban még nem tanulmányozták. Az általunk elvégzett farmakológiai szűrővizsgálatokban a növény kivonata markáns gyulladáscsökkentő hatást mutatott.
A pályázat keretében célul tűzöm ki a C. sadleriana kivonatának hatásorientált frakcionálását, a biológiailag aktív vegyületek izolálását és szerkezetük azonosítását, a kivonatok és a tiszta vegyületek gyulladáscsökkentő és sebgyógyulást elősegítő hatásának tanulmányozását. A növényi kivonat frakcionálását különféle kromatográfiás módszerekkel, a vegyületek szerkezetének meghatározását spektroszkópiás vizsgálatokkal végezzük el. A frakciók és tiszta vegyületek in vitro gyulladáscsökkentő (COX-1 és -2, 5-LOX-gátló hatás), in vivo sebgyógyulást segítő és gyulladáscsökkentő hatását tervezem tanulmányozni.
A munka további részében célul tűzöm ki további, Magyarországon honos, fitokémiai és farmakológiai szempontból kevéssé vagy egyáltalán nem vizsgált Centaurea fajok gyulladáscsökkentő hatásra történő szűrővizsgálatát és a jelentős aktivitású fajokkal részletes, a hatóanyagok azonosítására irányuló fitokémiai és farmakológiai munka elvégzését.
A tervezett kutatás hozzájárulhat egy, Magyarországon tradicionálisan alkalmazott gyógynövény hatásosságának igazolásához, a növény hatóanyagainak azonosításához. A kutatás tisztázhatja a gyulladáscsökkentő aktivitás szerepét a sebgyógyulás elősegítésében. A hazai Centaurea fajok gyulladáscsökkentő, sebgyógyító hatású vegyületeinek azonosítása értékes információkat szolgáltathat a gyógyszerkutatás számára és gyógynövény-eredetű termékfejlesztés alapjául szolgálhat.
Summary
The decoction of the aerial parts of Centaurea sadleriana, a plant native to Hungary, is traditionally used to treat the wounds of sheep in the Southern Great Plain region. Phytochemical and pharmacological studies of this plant have not been performed so far. Pharmacological screening of this plant carried out by our research group has revealed that the extracts of the plant possess a marked anti-inflammatory effect.
The proposed research programme aims the biological activity-guided fractionation of the extracts of C. sadleriana, the isolation and structure elucidation of its active compounds, and the investigation of anti-inflammatory and wound-healing effects of fractions and pure compounds. Fractionation of the plant extracts will be carried out by means of different chromatographic methods, and the structures of pure compounds will be elucidated by spectroscopic methods. The investigation of in vitro anti-inflammatory (COX-1, -2 and 5-LOX inhibitory), and in vivo wound-healing and anti-inflammatory effects of fractions and pure compounds is planned.
Other objectives of our planned research include the pharmacological screening of other, phytochemically and pharmacologically poorly or non-investigated Hungarian Centaurea species for anti-inflammatory effect and a detailed phytochemical and pharmacological work to identify biologically active agents from these species.
The proposed project may contribute to the confirmation of the efficacy of a Hungarian traditional medicinal plant, and to the identification of its active compounds. The investigation may reveal the role of anti-inflammatory activity in the wound healing effect. The identification of anti-inflammatory and wound healing compounds of Hungarian Centaurea species may provide valuable information for drug research and development. The results of this research programme may serve as a basis for the development of herbal medicinal products.

A Centaurea sadleriana Magyarországon őshonos, kizárólag a Kárpát-medencében előforduló faj. Föld feletti részéből készített főzetét tradicionálisan alkalmazzák haszonállatok sebeinek gyógyítására. Munkám célja a növény fitokémiai és farmakológiai vizsgálata volt, valamint a rokon fajok tanulmányozása a hasonló célú felhasználás szempontjából. A növény sebgyógyulást fokozó hatását állatkísérlettel igazoltam. Leghatásosabbnak a föld feletti rész apoláros, n-hexánnal készült kivonat bizonyult. A növény különböző polaritású oldószerekkel készült kivonatainak COX-1, COX-2 és 5-LOX enzimeket gátló hatását in vitro tanulmányoztam. Ezekben a tesztrendszerekben is az apoláros kivonatok mutatkoztak leghatásosabbnak. A kromatográfiás elválasztás hatáskövetésével azonosítottam a legaktívabb frakciókat, a fitokémiai munka elsősorban ezekre irányult. A munka keretében a C. sadleriana hatásorientált fitokémiai vizsgálata során 12 vegyületet izoláltam és azonosítottam a növényből különféle kromatográfiás technikák alkalmazásával, amelyek szerkezetét kromatográfiás módszerekkel határoztam meg. Tanulmányoztam 10, a Kárpát-medencében megtalálható rokon Centaurea faj in vitro COX-1, COX-2 és 5-LOX gátló hatását. A rokon fajok a C. sadlerianához hasonló mértékű hatással bírnak. Az esetleges gyógyászati felhasználás szempontjából a termesztésbe vont, illetve a vadon nagy mennyiségben megtalálható fajok tűnnek perspektívikusnak a kizárólag a Kárpát-medencében honos, védett C. sadlerianával szemben.

Centaurea sadleriana Janka is a species native Hungary, occurring only in the Carpathian Basin. Above-ground parts of the plant have been applied traditionally to cure the wounds of sheep int he Southern Great Plain in Hungary. The aim of my work was the phytochemical and pharmacological investigation of C. sadleriana and related species with potential similar medicinal application. The wound healing effect of the plant was proved in animal experiments. Apolar (n-hexane) extracts of the above-ground parts were the most active. The effect of different extracts on COX-1 and COX-2 and 5-LOX enzymes was studied in vitro. In these test systems the non-polar extracts appeared to be the most effective. Further activity-guided phytochemical investigation focused especially on the most active fractions. Int he course of this study, activity-guided phytochemical analysis of C. sadleriana using various chromatographic techniques resulted in the isolation and identification of 12 compounds, the structures of which were determined by spectroscopic methods. In vitro COX-1, COX-2 and 5-LOX inhibitory activities of 10 Centaurea species occuring in the Carpathian Basin were also studied. Thhese species exhibited similar activities to C. sadleriana. The potential therapeutic use of cultivated and more prevalent species appears to be promising.
