A 20 fehérjealkotó aminosav már viszonylag rövid oligopeptidek esetén is óriási variációs lehetőséget jelent. A nagy szám egyben sokféle, eléggé különböző tulajdonságú molekula. Ennek ellenére a Természet nem elégedett meg ezzel a variációs lehetőséggel és részben a riboszomális fehérjeszintézist követő poszttranszlációs módosításokkal, részben – főként növények, mikroorganizmusok, gombák által aminosavak módosításával nagyszámú, esetenként az anyavegyületre már alig hasonlító új vegyület jöhet létre. Ilyenek pl. a glikozilált, foszfatált stb. fehérjék és peptidek, de ilyenek a növényvilágban elterjedt, gyakorta gyógyszerként is felhasznált aminosav illetve peptid alapú heterociklusos vegyületek, az alkaloidok, az antibiotikumok egy része stb. A fehérjék poszttranszlációs módosításai alapvető jelentőségűek a különböző biológiai felismerési folyamatokban (szénhidrát felismerő lektinek, mannóz receptor az immunrendszer sejtjein, foszfotirozin felismerő SH2 és SH3 domének az immunrendszer regulációját biztosító fehérjéken, stb.). Mindezekért a fehérjealkotó aminosavakból felépülő polipeptidek szintézismódszereinek kifejlesztése után napjainkban egyre növekvő jelentősége van a módosított oligopeptidek szintézisének. Az eddigi jelentős fejlesztések komoly előrelépést jelentettek konformációsan gátolt illetve foszforilált peptidek szintézise terén, ugyanakkor még számos probléma részben vagy egészében megoldatlan. Pályázatunk keretében ezen problémák egy részének megoldását tervezzük. Ennek során az alábbi három fő területre koncentrálunk

1, Módszereket kívánunk kidolgozni ill. továbbfejleszteni – részben korábbi munkánk folytatásaként – különböző O és N glikopeptidek szintézisére. Ennek során szeretnénk összehasonlító vizsgálatokat végezni a synthon és a globális (konvergens) megközelítés hatékonyságával, valamint a felhasználható védőcsoportok és védőcsoporteltávolítási módszerekkel kapcsolatban.

2, Tovább kívánjuk folytatni a foszfopeptidekkel kapcsolatos vizsgálatainkat. Ennek során módszereket szeretnénk kifejleszteni enzimrezisztens foszfopeptid analógok ill. mimetikumok szintézisére, amelyekkel az immunrendszer regulációját biztosító fehérjék kölcsönhatásait tudnánk befolyásolni.

3, Vizsgálni kívánjuk a glikoziláció ill. foszforiláció hatásait a térszerkezeti viszonyokra, amely segítségével ezen peptidek és fehérjék hatásmechanizmusának megértéséhez juthatunk közelebb.
Summary
The number of possible combinations of the twenty proteinogenic amino acids is enormous even for smaller oligopeptides. However, Nature was not satisfied with this variability. Great amount of new compounds, hardly similar to the mother compound, can be formed by partly post-translational modifications following the ribosomal protein synthesis, partly modifications of amino acids by mostly plants, microorganisms and fungi. Beside glycosylated, phosphorylated, etc. peptides and proteins, such compounds are amino acid- and peptide-based heterocyclic compounds (alkaloids and some antibiotics, etc.) widespread in the plant kingdom and used as medicines. These post-translational modifications have fundamental importance in biological recognition processes. Because of these facts, after the development of synthetic strategies for the preparation of polypeptides containing proteinogenic amino acids, nowadays there is a growing importance of the synthesis of modified oligopeptides. As a result of intensive development in the past, major step forward has been achieved. However, numerous problems are partially solved or unsolved. We focus on the following three areas:

1, We plan to work out and improve methods – as continuation of our previous results - for the synthesis of O- and N-glycopeptides. We plan to do examinations in order to compare the effectivity of synthon and global (convergent) methods, and concerning protecting groups and their cleavage.

2, We plan to continue our examinations in connection with phosphopeptides. We plan to develop methods for the synthesis of enzyme resistant phosphopeptide analogues and mimetics that might affect the interaction of proteins insure the regulation of the immune system.

3, We plan to examine the effects of glycosylation and phosphorylation on the steric structure of peptides. Based on these, we might get closer to understanding mechanism of action of peptides and proteins.

Módszereket dolgoztunk ki oligo-szachariddal glikozilált N-glikopeptidek szintéziséremind konvergens, mind építőkő módszerrel. Védőcsoportkombinációkat optimalizáltunk O-glikopeptidek szintézisére, valamint optimalizáltuk a kapcsolási reakciót a glikán és a védett aminosav között. Számos foszfopeptidet és sejtpenetráló peptidet szintetizáltunk, részben fluoreszcensen jelzett formában is. Új foldamer molekulát (N-amino prolin) állítottunk elő és építettünk be béta-aminosav oligomerekbe. Megvizsgáltuk a kapott oligomerek térszerkezeti és stabilitásviszonyait.
Results in English
We improved the applicable methods for the synthesis of N-glycopeptides including convergent and building block approach. The protecting group combinations for the synthesis of O-glycopeptides was optimized and we improved the coupling conditions for the reaction of the glycosyl donor and acceptor. Numerous phospho and cell-penetrating peptide were synthesized and a part of them was fluorescently labelled. A new foldamer molecule (N-amino proline) was prepared and incorporated into beta-amino acid oligomers. The conformation and the stability of the new oligomers was investigated.
