Az egyes neuronok eltérő viselkedésének és válaszkészségének hátterében döntően az sejtfelszíni membránjukban található ioncsatornák és szinaptikus kapcsolataik különbözősége, valamint az azokat befolyásoló modulációs mechanizmusok eltérései állnak. Részben a kivételes morfológiai szituáció, részben előkísérletes adataink alapján kijelenthető, hogy a patkány nucleus cochlearis ideális modelljét kínálja a fenti folyamatok, összefüggések és kölcsönhatások tanulmányozásának. A jelen pályázatban négy fő kérdés vizsgálatát tervezzük: (1) Meg fogjuk határozni az ún. h-áram létrehozásáért felelős HCN alegységek típusát, megoszlását és életkorfüggését, mRNS, fehérje és funkcionális szinten. (2) Tanulmányozni fogjuk a mag egyes projekciós neuronjai által expresszált K+-csatornaalegységek aktivitásának és megoszlásának jelentőségét azok jellegzetes válaszreakcióinak kialakításában. (3) Meg kívánjuk határozni az olivocochlearis nyaláb által biztosított, funkcióját tekintve jelentős, ám részleteiben ezidáig nem tisztázott acetil-kolinerg kapcsolatok jelentőségét a neuronális válaszkészség és aktivitás befolyásolásában. (4) Bizonyítani kívánjuk, hogy a nucleus cochlearis Purkinje-szerű sejtjei valóban a cerebellum ectopiás Purkinje-sejtjei. A kérdések megválaszolását vékonyszelet-preparátumon végzett funkcionális mérések (a membránáramok- és potenciálok rögzítése, spontán és kiváltott szinaptikus aktivitások analízise), molekuláris biológiai (Q-PCR, Western-blotting) és immunhisztokémiai módszerek kombinált alkalmazásával tervezzük. Eredményeink elősegíthetik a sejtszintű jelfeldolgozás, valamint az egyes csatornaalegységek, receptormolekulák és modulációs mechanizmusok közötti összefüggések pontosabb megértését

The various response patterns and behaviours produced by individual neurones are mainly determined by their synaptic connections and ionic channels as well as by the different modulatory mechanisms affecting them. Based on a unique morphological situation present in the rat cochlear nucleus and on our preliminary data, this nucleus is an ideal model system for the investigation of the above processes, correlations and interactions. Four major questions will be examined in the present proposal: (1) We shall determine the types, distribution and age-dependence of the HCN subunits that are essential for the genesis of the h-current; the investigation will be performed at mRNA, protein and functional levels. (2) We shall examine the activity and distribution of several K+ channel subunits on certain projection neurones of the cochlear nucleus, affecting their unique neural activity patterns. (3) We shall study the properties and functional effects of the rather significant, yet hitherto not fully understood acetylcholinergic projections of the olivocochlear bundle on influencing the neural activity in the cochlear nucleus. (4) We wish to confirm that cochlear Purkinje-like cells are ectopic cerebellar Purkinje neurones. Answers for the above questions will be sought by a combined application of functional measurements (recording of the membrane potentials and membrane currents, analysis of spontaneous and evoked synaptic activity), molecular biology methods (Q-PCR, Western blotting) and immunohistochemistry techniques. Our results may contribute to a more thorough understanding of the correlations existing between neuronal signal processing, ion channel subunits, receptor molecules and modulatory mechanisms.

A projekt célja a nucleus cochlearis jelfeldolgozó tevékenységében jelentős ioncsatornák, modulációs mechanizmusok és szinaptikus kapcsolatok elemzése volt. A munka legfontosabb eredményei a következők: (a) Azonosítottuk a nucleus cochlearisban expresszált HCN-csatornaalegységeket. Feltártuk a h-áram jelentőségét az óriássejtek elektromos sajátságaiban és szinaptikus kapcsolataik finomhangolásában. (b) Leírtuk a kolinerg ingerlés hatását az óriássejtek elektrofiziológiai sajátságaira és a rajtuk kialakuló posztszinaptikus áramokra. Azonosítottuk a hatást közvetítő receptor-altípusokat. (c) Elemeztük a szemcsesejtek aktivitásfüggő intracelluláris kalciumkoncentráció-változásait. Kifejlesztettünk egy olyan félautomatikus kiértékelési eljárást, ami alkalmas a neuronális kalciumtranziensek felismerésére és analízisére. (d) Feltártuk egyes muszkarinerg receptorok szerepét sejttenyészeti körülmények között fenntartott astrocyták intracelluláris kalciumkoncentráció-változásaiban. (e) Képalkotó és elektrofiziológiai módszereket alkalmazva leírtuk a nucleus cochlearisban található Purkinje-szerű neuronok sajátságait. (f) Kvantitatív morfometriai analízis segítségével azonosítottuk a pyramis- és az óriássejtek elkülönítésére legalkalmasabb paramétereket. (f) Leírtuk a tacrolimus hatását a pyramis-sejteken kialakuló posztszinaptikus áramok tulajdonságaira. (g) Feltártuk a protein-foszfatáz-1M és a Rho-kináz szerepét a bushy-sejtek és az acusticus rostok közötti szinapszis működésében.

Our aim was to assess the roles of ionic channels, modulatory mechanisms, and synaptic connections in regulating the activity of the cochlear nucleus (CN). The most important results are as follows. (a) We identified all HCN channel subunits expressed in the CN. We revealed the significance of the h-current in shaping the electrical properties and fine-tunig the synaptic connections of the giant cells. (b) We described the effects of cholinergic stimulation on the membrane properties and postsynaptic currents of the giant cells along with the identification of the receptor subtypes mediating the effects. (c) We investigated activity-related cytoplasmic calcium concentration changes of the granule neurones, and developed a semiautomatic method for the identification and evaluation of neuronal calcium transients. (d) We assessed the involvement of various muscarinergic receptors in the regulation of intracellular calcium concentration changes of cultured astrocytes. (e) Using imaging and electrophysiological techniques, we described the properties of Purkinje-like cells of the CN. (f) We identified morphological parameters that are suitable for the unambiguous discrimination between pyramidal and giant neurones. (f) We documented the effects of tacrolimus on the postsynaptic currents recorded from pyramidal neurones. (g) We revealed and described the roles of the protein-phosphatase-1M and Rho-kinase in the synaptic transmission between the bushy cells and acoustic fibers.
