A biológiai rendszerek legfontosabb nyomelemei elsősorban a metalloenzimek aktív centrumának kialakításában, illetve sav-bázis és redoxi folyamatok katalizálásában vesznek részt. A réz-proteinekben a Cu(II)/Cu(I) átalakulás biztosítja a redoxi folyamatokban való részvételt. Ezeknek a rendszereknek a redoxipotenciálját a koordinálódó fémion geometriája, a kötődő donorcsoportok minősége jelentősen befolyásolja. Így a biológiai rendszerekben a réz-proteinek részvételével lezajló redoxi folyamatok még jobb megértését szolgálná a különböző réz(II)-peptid rendszerek redoxi sajátságainak feltérképezése. Így kutatási pályázatunk az imidazolgyűrű biológiai szerepének modellezésére szolgáló bisz(imidazol-2-il) vegyületek további átmenetifém-komplexeinek vizsgálata mellett kiemelt hangsúlyt fektetne egy szisztematikus ciklikus voltametriás vizsgálat sorozattal annak felderítésére, hogy a réz(II)ion-peptid rendszerekben a koordinálódó donorcsoportok helyzete és minősége milyen hatással van az képződő komplexek redoxi sajátságaira. Ez egyúttal arra is lehetőséget adhatna, hogy a különböző bisz(imidazol-2-il)csoportot tartalmazó származékok, valamint a SOD réz(II) kötőhelyét modellező több hisztidint tartalmazó tri- és pentapeptidek réz(II)komplexeinek redoxi sajátságairól is képet kapjunk, amely alátámaszthatja a feltételezett komplexek szerkezetét is, és esetlegesen információkat szolgáltathat a komplexekről, mint lehetséges enzim modellekről.
Summary
Trace elements of biological systems take part in the formation of the active centre of metalloenzymes, and catalyze different acid-base and redox reactions. For example, Cu(II)/Cu(I) systems serve as electron transfer in the redox reactions catalysed by copper-metalloproteins. The redoxpotential values of these systems depend on the coordination geometry of the metal ion and the type of donor atoms. Investigation of redox properties of copper(II)-peptide systems may provide valuable information on the redox processes connecting to copper-metalloproteins. The main goal of our research project is the systematic study of copper(II) and dipeptide systems containing different side chain donor atom by cyclic voltammetry. These results may give possibility to understand the influence of the type of donor atoms and coordination geometry of complexes for their redox properties. The cyclic voltammetric studies of copper(II) complexes of bis(imidazol-2-yl) derivatives and multihistidine containing tri- and pentapeptides may result in new evidences for the structure of complexes supposed on the base of potentiometric and spectroscopic parameters, and may give more information about complexes as possible enzyme models.

1. A Cu(II)-aminosav- és -peptid rendszerek elektrokémiai vizsgálatból az a következtetés vonható le, hogy a hisztidin imidazol-oldalláncának van a legnagyobb hatása a komplexek redoxipotenciál értékére. Az (NH2,Im(N)) koordinációval keletkező komplexek réz(II)-centrumának redukálhatósága jelentős mértékben növekszik. A réz(II)-multihisztidin-peptid rendszerekben a 2, 3, 4 imidazol-koordinációjú mono-, illetve bisz-komplexekre általánosan a pozitív redoxipotenciál érték volt a jellemző, melynek értéke csökken az imidazol-N számának növekedésével, a komplex stabilitásának növekedésével. A jóval merevebb bisz(imidazol-2-il) koordinációjú komplexek redoxipotenciál értékei 0 V körüli értékek. 2. A legígéretesebb komplexek SOD-aktivitásának meghatározásából az következett, hogy a fémionhoz kötődő hisztidin imidazol nitrogének számának növekedésével az aktivitás növekedik, így a legnagyobb aktivitást a négy hisztidint tartalmazó peptid esetén kaptunk. Ugyancsak jelentős mértékű SOD aktivitás figyelhető meg az imidazolátohidas bisz(imidazol-2-il)-komplexek esetén. 3. Számos aromás és heteroaromás ligandum vanádium(IV), vas(III), vas(II) komplexeinek oldategyensúlyi és elektrokémiai vizsgálata azt mutatta, hogy ezek a fémionok kisebb stabilitással kötődnek az aromás-N donoratomokhoz, viszont kiugró stabilitású (O–,N,O–) koordinációjú komplexek képződését mutattuk ki aromás hidroxi és N-donoratomot tartalmazó ligandumok esetén. Ezek potenciális vas-kelátorok lehetnek.
Results in English
1. The electrochemical measurements of the Cu(II)-amino acid and peptide systems showed that the imidazole chain of histidine has the greatest effect on the redox potential value of the complexes. The reducibility of the Cu(II) center of complexes with (NH2, Im(N)) coordination increases significantly. In the Cu(II) multihistidine peptide systems the redox potential values for the mono and bis complexes with 2, 3, 4 Im(N) coordination were positive generally. This value decreased by the increase of the number of Im(N) thus with the increase of the stability of the complexes. The redox potential values of the complexes with more rigid bis(imidazolyl) type coordination are around 0 V. 2. The measurement of the SOD activity of the most promising model complexes reflected that the activity increases with the increase of coordinated Im(N). The greatest activity was measured for the complex of the four histidine containing peptide. An equally significant SOD activity can be observed for the imidazolato bridged bis(imidazolyl) complexes. 3. The solution equilibrium and electrochemical studies of the VO(IV), Fe(III) and Fe(II) complexes of several aromatic and heteroaromatic ligands showed that these metal ions bind to the aromatic-N donor atoms with smaller stability. However, the formation of (O–,N,O–) coordinated complexes with outstanding stability was proved in the case of ligands containing aromatic hydroxy group and N-donoratoms. These can be potential iron chelators.
