A fotoreceptor-fejlődés sejt- és molekuláris biológiája

A szembe érkező fényingereket a retina fotoreceptor sejtjei, a csapok és a pálcikák alakítják ingerületté. Ezek kültagjaiban találhatóak az ún. fotopigmentek, amelyek a jelátviteli folyamatot beindítják. Az alacsony ingerküszöbű, rodoposzin tartalmú pálcikák a szürkületi fényre is érzékenyek, sőt fel is erősítik azt, míg a csapok a nappali látásért felelősek.
Az elmúlt évek számos közleménye arra utal, hogy a fototranszdukció résztvevői egy közös speciális membránterületen, ún. lipid rafton lelyezkednek el A lipid raftoknak egy speciális alcsoportját képezik a caveolák, melyek egy jellegzetes intramembrán proteinnel, caveolinnal rendelkeznek Az elmúlt években vizsgálatokat végeztünk azzal kapcsolatban, hogy a retina tartalmaz-e caveolinokat. Elsőként írtuk le az emberi retina, később az örvösnyakú maki retinájának caveolin eloszlását. Újabban az érdeklődés középpontjába került, hogy a caveolin-1-nek fontos szerepe van a fejlődés során. Ennek hatására kezdtünk el vizsgálatokat végezni a fejlődő retinákon. Előzetes eredményeink szerint mind a caveolin-1, mind a cavelon-2 expresszálódik a fejlődés során, sőt az expressziójukban változások figyelhetőek meg. Vizsgálataink során a hagyományos morfológiai technikákat (fénymikroszkóp, elektron mikroszkóp, konfokális mikroszkóp) szeretnénk ötvözni a legújabb biokémiai módszerekkel (Western blot, immunprecipitáció, pcr, Fret analízis). Ezek segítségével kívánjuk végigkövetni a fototranszdukciós fehérjék expresszióját a fejlődés során és választ találni arra a kérdésre, hogy mi a caveolinok pontos szerepe a retinában ill. a fotoreceptor sejtekben.
A téma sokszínűsége és újszerűsége miatt meggyőződésünk, hogy eddigi előzetes eredményeink csak bevezető lépései egy igen nagy horderejű témának, amely nem csak az alapkuatatásban kaphat helyet, hanem később klinikai kuatatások alapjául szolgálhat

Cell- and molecular biology of photoreceptor development

The process of visual perception begins in the retinal photoreceptor cells: the rods and the cones. The rods, containing rhodopsin, have lower threshold sensitivity; they amplify light signals more than cones, and are therefore responsible for twilight (scotopic) vision. Recent studies suggest that the phototransduction members and also some of their modulators are arranged in special membrane domains, called lipid rafts. Detergent-resistant membranes (DRMs) have been isolated as in vitro models of lipid rafts, from photoreceptor outer segments. Photoreceptor DRM are also enriched in caveolin-1, a protein consituent of specialized lipid raft domains called caveolae. Only a few reports have been published about the presence and distribution of caveolin in the retina so far. We were the first to report the distribution of caveolin isoforms in the retina of human, then in the ruffed lemur. However the expression of caveolin-1 and caveolin-2 in rods is no longer a matter of dispute, their exact function is still unanswered.
Lately it has been demonstrated, that caveolin-1 plays an important role in neural development, which influenced us to start observations about the caveolin distribution in developing retinas. We found that both caveolin-1 and caveolin-2 are expressed in the developing retinas and additionally their expression level seem to change during the development. To understand the expression, rearrangement and modulation of phototransduction members we would like to combine the classical morphological methods (light microscopy, confocal microscopy, electon microscopy) with the modern biochemical ones (Western blot, immunoprcipitation, pcr, Fret analysis). We are sure, that our preliminary results are only a the first few steps in a broad topic, in which efficient work is expected. Our prospective results can not only broaden facts in basic reseach, but also can be basis for clinical studies.

Az opszinváltás szabályozása fajonként eltérő, tiroxin és TRβ2 nélkülözhetetlen a zöld csapfejlődéshez. A receptormegoszlás időbeli és térbeli mintázatával igazoltuk a tiroxint mediáló TRβ2 szerepét. Tenyésztő módszerünkkel a fotoreceptor-differenciálódás szérummentes környezetben is végbemegy, a világon először definitív médiumban vizsgálható. Más faktorok (A- és E-vitamin, BDNF) is hatnak a pigment-expresszióra. Szelektív neurotrophin antagonisták az opszin és a TrkB receptor kapcsoltságára utalnak. Az erythropoietin retinális expressziója HIF1-α szabályozás alatt áll és időben változik a fejlődés korai szakaszában. Szemnyitás után a retinában lecsökken a transzdukciós molekulák szintje, kivéve a fotoreceptorokat. A sejttestben szintetizálódnak, közös lipid rafton kerülnek a kültagba és játszanak szerepet a fejlődésben. Az STK38L gén homozigóta mutációja befolyásolja a fotoreceptor-fejlődést. Sejthalál és proliferáció egyaránt jelen van. Hibrid sejtek jelennek meg pálcika-, kisebb mértékben S-opszin termeléssel. A differenciált, mutáns sejtek a degenerációs gén hatására megőrzik osztódóképességüket. A melatonin éjszaka termelődik, éjjeli világítás a hormontermelés gátlásával patológiás folyamatokat (emlő és colorectalis carcinoma) okozhat. A pineális szerv emlősben elvesztette fotoreceptor működését és szimpatikus rostokon a retinából kap információt. Mivel a pineális melatoninképzést rövidhullámú fény gátolja, éjjeli műszakban hosszúhullámú megvilágítást kell használni.

Regulation of opsin-switch varies across species. Thyroxin and TRß2 receptor is essential for green cone development. Spatial and temporal receptor distribution proved the role of TRß2 mediating thyroxin. Photoreceptor differentiation is completed in our culture method in serum-free medium, allowing its examination in a definitive paradigm. Other factors such as Vitamins A and E, BDNF) also influence visual pigment expression. Selective neurotrophin antagonists reveal the close connection of opsin and Trkß. Retinal expression of erythropoietin is under the regulation of HIF1- α, and changes during development. Transduction molecule levels decrease after eye opening, except for photoreceptors. They are synthesized in the cell body, located and transported in the outer segment on common lipid rafts and play a developmental role. Homozygotic mutation of STK38L influences photoreceptor development. Cell death and proliferation are equally present. Hybrid photoreceptors appear that express rod and, to a lesser extent, S-opsin. The degenerated, hybrid mutants retain their capacity to divide. Melatonin is produced at night. Nocturnal light exposition inhibiting hormone production may generate pathological processes (e.g.: carcinomas). The pineal organ in mammals has lost its photoreceptor function and receives information from the retina via sympathetic fibers. Since short wave light inhibits melatonin production, long-wave illumination is recommended during night-shift.
