A súlyosan idült neurodegeneratív betegségek az agy különböző területein lévő neuroncsoportok szelektív pusztulásának következményei. A „rossz” szerkezetű fehérjékből aberráns fehérje-fehérje kölcsönhatások során fehérjeaggregátumok és zárványtestek képződnek. A strukturális/ultrastrukturális változásokat intenzíven tanulmányozzák. Azonban kevéssé ismertek az ezekkel a folyamatokkal együtt járó enzimatikus és metabolikus változások, habár a mitokondrium működési zavarát és az energiatermelés károsodását is olyan mechanizmusnak tekintik, melyek szerepet játszhatnak a neurodegeneráció kialakulásában. Ezen kutatás célja az energia metabolizmus lokális és rendszerszintű jellemzése a Huntington (HD) és a Parkinson (PD) kór állat és sejt modelljeiben. A kísérleteket transzfektált humán sejtvonalakon, neurotoxinnal kezelt egerek agyszöveti mintáin, és a HD esetében transzgenikus egerek agyszöveti mintáin is, tervezem végezni. Vizsgálataim azt célozzák, hogy a neurodegenerációs betegségekre jellemző fehérjék jelenléte és az ennek következtében kialakuló patológiás változások milyen hatást gyakorolnak a glikolízisre és a mitokondriális elektron transzport rendszerre. A biológiai modellezés továbbá lehetővé teszi az energia metabolizmus kvantitatív jellemzését fiziológiás és patológiás körülmények esetén.
A projekt sikeres teljesítése nagymértékben hozzájárul ahhoz, hogy megértsük az energia metabolizmusban bekövetkező változások szerepét a neurodegenerációs folyamatok kialakulásában, továbbá elősegíti potenciális drog-célpontok azonosítását, ami elősegítheti a neurodegenerációs betegségek kezeléséhez alkalmas gyógyszerek fejlesztését.
Summary
The progressive neurodegenerative diseases are caused by the selective loss of neurons in various areas of brain. Unfolded/misfolded proteins participate in aberrant protein-protein interactions resulting in aggregation and plaque formation. The structural alterations have been the object of extensive studies. However, the concomitant enzymatic and metabolic alterations are still unclear, although mitochondrial dysfunction and energy impairment have been suggested as mechanisms underlying the pathogenesis of neurodegeneration. The aim of this project is to characterize and analyze systematically the energy metabolism at local and at system level in mouse and cellular models of Huntington’s (HD) and Parkinson’s (PD) disease. For the experiments transfected human cell lines and neurotoxin treated mice will be used, in the case of HD a transgenic mouse model also will be investigated. The effect of pathological alterations due to the presence of different hallmark proteins of neurodegeneration on glycolysis and on mitochondrial electron transport system will be investigated, and the energy metabolism at physiological and pathological situations will be quantitatively characterized by means of biosimulation.
The successful fulfilment of this project will significantly contribute to the understanding of the role of alterations in energy metabolism related to the pathomechenism of HD and PD. It may assist to identify potential molecular targets for developing therapeutic agents for the treatment of these neurodegenerative disorders.

A neurodegenerációs betegségek (Alzheimer-, Parkinson-, Huntington-kór) patomechanizmusának tisztázása napjaink kiemelt kutatási témája. Jellemeztük az e betegségek kialakulásában kulcsszerepet játszó fehérjék kölcsönhatásait és az energiametabolizmusra (glikolízis, mitokondriális komplexek, ATP-szint) gyakorolt lokális és rendszer szintű hatásait; valamint egy új, alfa-synucleinnel ko-lokalizáló fehérje (TPPP/p25) fiziológiás és patológiás hatásait vizsgáltuk. A rendezetlen vagy mutáció következtében megváltozott szerkezetű fehérjék aberráns kölcsönhatásaiban kis, toxikus oldható oligomerek játszanak szerepet. Az egyedi fehérjék strukturális és funkcionális sajátságai a különböző ultrastruktúrák kialakulása révén megváltoznak; pl. az alfa-synuclein/beta-amyloid gátolja a TPPP/p25 mikrotubulus szervező aktivitását. A TPPP/p25-beta-amyloid kölcsönhatás kevert típusú neurodegenerációs betegségekben játszik szerepet. Stratégiát dolgoztunk ki ezen fehérjék energiametabolizmusra gyakorolt hatásainak molekuláris szintű jellemzésére. Az egyedi enzimek sebességi egyenletein és kísérletesen mért kinetikai paraméterein alapuló matematikai modell kidolgozása lehetővé tette a lokális változások rendszer szintű megnyilvánulásainak jellemzését Huntington-kór modellekben és a TPI deficienciában. Eredményeink hozzájárulnak a neurodegenerációs betegségek patomechanizmusának jobb megismeréséhez, ami mind a diagnosztika, mind pedig a gyógyszercélpontok azonosítása szempontjából jelentős.

Nowadays research on the pathomechanisms of neurodegenerative diseases (Alzheimer’s, Parkinson’s, Huntington’s diseases) is of special importance. We investigated the interactions of proteins playing key role in the development of these diseases, their effect on energy metabolism (glycolysis, mitochondrial complexes, ATP-level) at local and system levels; and the physiological and pathological roles of a new protein (TPPP/p25) enriched in pathological inclusions. The unfolded or misfolded (due to mutation) proteins form aberrant interactions, in which small, toxic soluble oligomers play crucial role. The formation of different ultrastructures alters the structural-functional characteristics of these proteins; e.g. the interaction of TPPP/p25 with alpha-synuclein/beta-amyloid impedes its tubulin polymerization promoting potency. The TPPP/p25-beta-aymloid interaction may play a role in mixed type neurodegenerative diseases. To investigate the effects of hallmark proteins on the energy metabolism, the following strategy was applied. A mathematical model based on the experimentally determined kinetic parameters and the rate equations of the glycolytic enzymes was elaborated to investigate the effects of local alterations at system level in Huntington’s disease models and TPI deficiency. Our results contribute to the better understanding of the pathomechanism of neurodegenerative diseases, which is important both in diagnostics and identification of possible drug targets.
