Sokáig bevett nézet volt, hogy minden fehérjéhez rendelhető egy egyedi úgynevezet natív szerkezet, amely szükséges és elengedhetetlen a fehérje működéséhez. Az utóbbi években azonban számos rendezetlen, szerkezet nélküli fehérjét (Intrinsically Unstructured Protein, IUP) írtak le, amelyek bár specifikus biológiai funkciókban vesznek részt, nem rendelkeznek egyedi, jól definiált szerkezettel. A jelen munkában az IUP-k és a rendezett fehérjék közti termodinamikai különbségek felderítését tűztük ki célul. A két legfontosabb termodinamikai paraméter, nyomás és hőmérséklet változtatása mellett érzékeny spektroszkópiai módszerekkel követjük a fehérjék szerkezeti változásait.
Model fehérjének a titin óriásfehérje PEVK doménjét választottuk, amelyet a szakirodalom prototipikus IUP-nek tart. A PEVK számos rekombináns fragmentumában vizsgáljuk, hogy indukálható-e lokális szerkezet, és különböző kísérleti paraméterek ezt hogyan modulálják.
Céljaink és feladataink az alábbiak: Indukált lokális és globális szerkezetek detektálása IUP-ban. Ezen indukált szerkezetek és a természetes (rendezett) fehérjeszerkezetek stabilitásának összehasonlítása. Kétértékű ionok által a PEVK-ban indukált lokális rendezett formák jellemzése. A PEVK Ca ion kötő régiójának azonosítása. Az indukált és természetesen előforduló szerkezetek mechanikai stabilitását nagy nyomás perturbáló hatásán keresztül vizsgáljuk. Megmérésre kerül az indukált szerkezetek kialakulási kinetikája mind a rendezetlen, mind pedig természetesen rendezett fehérjék esetében.

For many years it has been thought that every protein has its unique structure (folded or native conformation) necessary for its function. Recently, however, a number of intrinsically disordered or unstructured proteins (IUPs) have been described. Although IUPs do not possess unique, well defined structures, they can be associated with specific biological functions. In this work we aim to reveal the thermodynamic differences between IUPs and ordered proteins. We plan to modulate the two most important intensive thermodynamic parameters, temperature and pressure, and follow the structural changes of the proteins by highly informative spectroscopic methods. As a model protein to investigate we selected the PEVK domain of the giant protein titin, which has long been considered a prototypical IUP. Specific fragments of the PEVK domain will be expressed and used for analyzing how local structure might be altered and induced by a variety of experimental factors.
Our specific aims and tasks are the following: Detection of induced local and global structures in disordered proteins. Comparison of stability of the induced structures with the stability of naturally ordered proteins. Characterization of local structures in PEVK polypeptides by binding of divalent ions. Identification of the Ca ion binding region of the PEVK domain.We plan to use of high-pressure perturbation for detection of mechanical stability of the induced and naturally occurring structures. Kinetic characterization of the induced structural changes in the disordered as well as in the ordered proteins will be achieved in pressure jump experiments.

A projekt célja a nemrégiben leírt rendezetlen fehérjék (RF) és a rendezett fehérjék összehasonlítása volt, változatos fizikai és kémiai körülmények között. Széles hőmérsékleti és nyomástartományban, ill. különböző kémiai ágensek, kozmotróp anyagok jelenlétében vizsgáltuk, hogy a RF-kben lehet-e lokális v. globális szerkezetet indukálni. A már rendezett fehérjék esetén a rendezett szerkezet p-T fázisdiagramjának meghatározása volt a feladat. A következő fehérjéket, ill. fragmentumaikat vizsgáltuk: Titin PolyE, PEVK I, PEVK 11 és PEVK 21 fragmentumai, titin I27 domén, parvalbumin, lizozim L99A mutáns, Mal d 1 és az Rv3221c fehérjék, foszfoglicerát kináz, poliglutaminsav, ill. poliaszparaginsav. A titin rendezetlen doménjét ugyan nem sikerült fizikai és kémiai környezet változásával globálisan rendezetté tenni, azonban így is sok értékes információt tudtunk meg ezeknek a különböző méretű rendezetlen fragmentumoknak a viselkedéséről széles környezeti paramétertartományban. A rendezett fehérjéket illetően meghatároztunk számos nyomás-hőmérséklet fázisdiagramot. Megállapítottuk, hogy az Rv3221c fehérjét hibásan sorolták a rendezetlen fehérjék közé, meghatároztuk a fázisdiagramját és a hozzá tartozó termodinamikai paramétereket. Tudományos eredményeinket összesen 8 referált folyóiratcikkben és egy könyvfejezetben publikáltuk, valamint számos konferencián ismertettük. A már publikált eredmények mellett öt másik publikáció van előkészületben.

The aim of the project was the comparison of the recently described intrinsically unstructured proteins (IUP) with ordered proteins, under various physical and chemical parameters. We investigated, whether nonphysiological temperature, pressure range, or cosmotropes or other chemical agents can induce locally or globally ordered structures. In case of ordered proteins the determination of the p-T diagram of the proteins was the main aim. The following proteins and fragments were investigated: PolyE, PEVK I, PEVK 11 and PEVK 21 fragments and the I27 domain of titin, parvalbumin, lysozyme L99A mutant, Mal d 1 and Rv3221c proteins, phosphoglycerate kinase, polyglutamic acid and polyaspartic acid. Although various chemical, and physical parameters were varied, globally ordered structures were not induced in the titin fragments, but a series of valuable information was obtained about the behavior of these fragments in wide pressure and temperature regions. We determined a number of pressure-temperature phase diagrams. We proved that the Rv3221c protein was not an IUP, and determined its phase diagram, and the corresponding thermodynamical parameters. The results were published in 8 referred articles and in a book chapter. Besides, we presented these in a number of conference publications. Five papers are still in preparation.
