A nitrogén-monoxid (NO) fontos szerepet játszik a növény számos élettani és fejlődési folymataiban, beleértve a gyökér architektura kialakulását is. Jelen publikált munkánkban az ozmotikus stressz és a külsőleg adott auxin (indol-3-vajsav, IBA) hatását vizsgáltuk borsó növények gyökér fejlődésére és az NO produkciójára. A NO detektálást in vivo és in situ végeztük fluoreszcenciás módszer alkalmazásával. Növekvő polietilén glikol (PEG) és IBA koncentrációk hatására az elsődleges gyökerek hossza csökkent, viszont megemelkedett a az oldalgyökerek száma, valamint a NO szintje. Ozmotikus stressz alatt az NO képződés időbeni kinetikája jelentősen különbözött az auxin jelenlétében észlelt kinetikától, ugyanis a lateráris gyökériniciálisok megjelenését erőteljes, tranziens NO-képződés előzte meg. Feltételeztük, hogy ez a korai NO-megjelenés az ozmotikus stressz alatti jelátvitelben játszik szerepet, amely út a gyökér morfológiájának megvátozásához vezet. A NO keletkezési módja a különböző növényi válaszreakciókban nem teljesen ismert. Arabidopsis mutánsok segítségével kimutattuk, hogy az auxin jelenlétében indukálódó oldalgyökér képződéskor a NO keletkezése a nitrát-reduktáz (NR)aktivitásához kapcsolható, amint azt a NR-deficiens nia1,nia2 és az Atnoa1 (korábban Atnos1) mutánsok reakciói bizonyították. Kimutattuk, hogy az IBA NO-képződést indukált mind a vad típusú, mind pedig a NO-szintáz szerű enzim hiánymutánsban, viszont a NO teljesen hiányzott a NR hiánymutánsaiban. Az oxidatív stressz alatt indukálódó korai NO csúcs eredete az auxin-indukálta NO-tól valószínűleg különbözik.
Summary
Nitric oxide (NO) functions in variable physiological and developmental processes in plants, including the establishment of root architecture. In this work the effects of osmotic stress and exogenous auxin (indole-3-butyric acid, IBA) on root morphology and nitric oxide (NO) generation in roots were compared in pea plants. NO generation was examined by in situ and in vivo fluorescence method. Increasing concentrations of polyethylene glycol (PEG) as well as IBA resulted in shortening of primary root (PR), enhancement of lateral root (LR) number and significant increase of NO generation. Under osmotic stress, the time curve of NO development was distinct compared to that of IBA-treated roots, since significantly, the appearance of lateral initials was preceded by a transient burst of NO. This early phase of NO generation under osmotic stress was clearly distinguishable from that which accompanied LR initiation. We suppose that the early phase of NO generation may fulfill a role in the osmotic stress-induced signalization process leading to the modification of root morphology. However, the source of NO in the diverse plant responses is not well understood. We provide genetic and pharmacological evidence that the production of NO is associated with the nitrate reductase (NR) enzyme during IBA-induced lateral root development in Arabidopsis thaliana L. For investigation this question, we used the NR-deficient nia1, nia2 and Atnoa1(former Atnos1) mutants of Arabidopsis. As inhibitor for NO synthase (NOS) NG –monomethyl-L-arginine (L-NMMA) was applied. It was clearly demonstrated that IBA was able to induce NO generation in both the wild type and Atnoa1 plants, but it totally failed in the NR-deficient mutant. It is concluded that the source of NO is the nitrate reductase enzyme during IBA-induced lateral root development in Arabidopsis thaliana.
