A prion betegségek halálos neuro-degeneratív működési zavarok. Az emberi szervezetre igen veszélyesek, mivel mind a táplálékláncon mind orvosi beavatkozás alkalmával átvihetők. Jelen kutatási tervben (1) az egészséges prion proteint és annak mutáns változatait vizsgáljuk azzal a céllal, hogy felderítsük a mechanizmust, amely a TSE előre haladtával az idegsejtek pusztulásának hátterében áll. Továbbá, (2) egy új megközelítést alkalmazva olyan szegmenseit azonosítjuk a prion fehérjének, amelyek másodlagos szerkezete átalakul béta konformációvá az abnormális, proteáz-rezisztens prion forma (PrPres) keletkezése során. Mindkét kérdés alapvető fontosságú annak érdekében, hogy a TSE kezelésében megfelelően érzékeny diagnosztikai módszereket és hatékony terápiát lehessen kidolgozni.
Az első kérdést különösen jelentőssé teszi, hogy mostanában mutatták ki, hogy oldható oligomerjeit az amyloid-béta proteinnek (amelyet az Alzheimer betegségben a sejtekre toxikusan ható formának tekintenek) a sejtek a celluláris prion fehérjén keresztül veszik fel és PrP részt vesz hatásának a közvetítésében.
Summary
Prion diseases are deadly, neuro-degenerative disorders. They are especially dangerous, since they can be transmitted to humans through the food chain or by medical intervention.
Here, (1) we explore the normal healthy prion protein and its mutant forms to reveal the mechanism of the neuronal cell death occurring during the progression of TSE. Furthermore, (2) by using a novel approach, we aim to identify the segments of PrP that undergoes a conformational change to beta-strand during the conversion to PrPres.
Both issues pose fundamental questions in the field and are critical for developing sensitive diagnostic methods and an effective therapy for TSE.
The first issue has become particularly relevant by the recent finding that the cellular form of PrP mediates the uptake and effects of the oligomeric form of amyloid-beta protein that is considered as the toxic entity to the neuronal cells in Alzheimer’s.

Kutatómunkánk célja megérteni a prion fehérje és a prion-fehérje család fiziológiás funkcióját és szerepét neurodegeneratív betegségekben, a szivacsos agysorvadás és az Alzheimer-kórban. Elsősorban a fehérje neurotxikus és neuroprotektív hatásait, illetve neurogenezisben betöltött szerepét próbáljuk megérteni. Továbbá, a szivacsos agysorvadás betegségek során keletkező kóros konformációjú prion fehérje szerkezetéről és kialakulásának mechanizmusáról próbálunk pontosabb képet kialakítani. Új módszereket állítottunk be és közöltünk tudományos folyóiratokban, amelyek lehetővé teszik sejtkultúrában is vizsgálni a fenti kérdéseket. Megállapítottuk, hogy a prion fehérjék neurotoxikus potenciálja az eddigi elképzelésektől eltérően, a fehérjék rendezetlen N-terminális részeihez köthető. Kísérleteink megmutatták, hogy Retinsavval indukált idegsejtekké differenciálódásuk során a prion fehérje expressziója nagy mértékben nő, míg ugyanez a prion fehérje család többi tagjáról nem mondható el. Azonosítottunk egy szegmenset a fehérjében, ami a fehérjedimer kölcsönható felszínének a része. -- A prion fehérje kóros konformációjú formája aggregált, multimer állapotú. -- Amennyiben a jellemzett dimerek a multimer képződés útvonalának egyik állomását jelentik, úgy ez a kölcsönható felszín fontos gyógyszer
Results in English
The purpose of our work is to understand the physiological function of the prion protein and the prion protein family as well as their role in the pathomechanism of transmissible spongiform encephalopathy and Alzheimer disease. We mainly focus on the neurotoxic and neuroprotective functions of the prion protein family and on its role in neurogenesis. We also examine the structure and the mechanism of formation of conformationally altered variants of the prion protein that are typical in the progression of TSE. We have developed new methods published in scientific journals, facilitating to interrogate the above mentioned questions in cell culture systems. In our experiments, in contrast to the present view, the neurotoxic potential of the prion proteins is associated with the natively unfolded N-terminal parts of the proteins. Examining the expression levels of the members of the prion protein family during retinoic acid induced neuronal differentiation, we revealed that only the prion protein shows greatly increased expression. We identified a segment in the dimer interface of the prion protein. -- The disease-associated form of the prion protein (PrPSc) is aggregated, multimeric in nature. -- As much as this dimeric form is an on-pathway intermediate on the way to the formation of the PrPSc, the interacting surface identified here is an important drug target.
