Korábbi munkánk során igazoltuk, hogy a kóros májszövet az éphez képest jóval nagyobb mennyiségű, és megváltozott biológiai aktivitású glükózaminoglikánt tartalmaz. A jelen kutatás során a fibrogenezissel járó krónikus májbetegségekben és a májrákban jellemeztük a heparánszulfát szerkezeti és mennyiségi változásait, valamint a heparánszulfátot módosító enzimek expresszióját. Immunhisztokémiai, kvantitatív RT-PCR és diszacharid-elemzési vizsgálataink azt mutatták, hogy ezekben a kóros állapotokban a heparánszulfát felhalmozódásával egyidejűleg megnő a heparánszulfát módosítását végző enzimek expressziója, továbbá megváltozik a heparánszulfát szulfatáltságának mértéke és mintázata. A 3-O-pozícióban fokozott, míg a májrákban a 6-O-pozícióban csökkent szulfatálást tapasztaltunk. Mivel a heparánszulfát kölcsönhat az extracelluláris mátrix növekedési faktoraival, és részt vesz a májgyulladást okozó vírusok megkötésében, eredményeink adalékokat szolgáltatnak a krónikus májbetegség egyes kórfolyamatainak jobb megértéséhez.
Summary
Previously, we have demonstrated markedly elevated levels and altered biological activity of glycosaminoglycans in the chronically diseased liver tissue. In the present work we analyzed the quantitative and structural alterations of heparan sulfate (HS), as well as the expression of heparan sulfate modifying enzymes, in fibrogenic liver diseases and hepatocellular cancer. Based on our immunohistochemical, quantitative RT-PCR and disaccharide analysis experiments we concluded that in parallel with the accumulation of HS in these pathological conditions, the expression of HS modifying enzymes is also increased. Moreover, we have revealed an altered pattern and extent of HS sulfation: whereas 3-O-sulfation was found to be increased in all disease conditions, 6-O-sulfation was slightly diminished in liver cancer. Since HS is known to interact with growth factors in the extracellular matrix, and acts as a cofactor in the binding of hepatitis viruses, our results may contribute to a better understanding of certain pathomechanical aspects of chronic liver disease.

A heparánszulfátok fontos szerepet játszanak a sejtek közötti kommunikációban, a jelmolekulák hozzáférhetőségének és hatékonyságának szabályozásában, így élettani és patológiás körülmények között is számtalan folyamatot befolyásolnak. Kimutattuk, hogy e molekulák szerkezete és elhelyezkedése jellegzetes módon megváltozik a máj krónikus betegségeiben (jó- és rosszindulatú májdaganatokban és májzsugorban), és hogy e változások összefüggésbe hozhatók a módosult enzim-expressziókkal. Végezetül rámutattunk e változások néhány implikációjára a betegségek mechanizmusával és esetleges kezelésével kapcsolatban.

PubMed: J Histochem Cytochem. 2010 May;58(5):429-41. Epub 2010 Feb 1. PMID: 20124094 In the paper we examined heparan sulfate (HS) glycosaminoglycans and their modifying enzymes in the liver. HS plays pivotal roles in intercellular communication by regulating the availability and effectiveness of signal molecules. Thus, HS modulates a vast array of physiological and pathological processes. We demonstrated that both the structure and localization of HS undergoes specific alterations in chronic liver diseases including benign and malignant tumors as well as cirrhosis. These alterations were related to changes in the expression of various HS-modifying enzymes. Finally, we tried to interpret our findings in the light of pathomechanism and future therapies.
