A defenzinek és a hasonló antimikrobiális proteinek széles körben elterjedtek az élővilágban: jelenlétüket eddig öt fonalasgombában is kimutatták. A rendszertanilag távol álló fajok termelte, szerkezetileg hasonló proteinek hatásmódjában és fajspecificitásában jelentős eltérés mutatkozik. A Neosartorya fischeri antimikrobiális protein (NFAP), a N. fischeri (anamorfja: Aspergillus fischerianus) genomjában megtalálható szekvenciából származtatott hipotetikus antifungális protein, jelenléte csak in silico bizonyított. Klónozásáról, izolálásáról, antifungális hatásspektrumáról, hatásmódjáról és biológiai szerepéről még nem számoltak be. A pályázat célja, hogy bővítsük a fonalasgombák által termelt defenzin-szerű antimikrobiális proteinekről meglévő ismereteinket az NFAP tanulmányozásával.
Ezek alapján kutatási tervünkben a következő célkitűzéseket fogalmaztuk meg: (1) az NFAP-t kódoló gén és ortológjainak azonosítása egy Neosartorya fischeri (NRRL 181) izolátumból és rokon fajokból; (2) az NFAP antimikrobiális tulajdonságainak és hatásmódjának vizsgálata; (3) heterológ expressziós rendszer létrehozása az NFAP termelésére; (4) az NFAP szerkezete és hatásfoka közötti összefüggés feltárása; (5) az NFAP biológiai szerepének a vizsgálata.
Eredményeink adatot szolgáltatnának a defenzin-szerű antimikrobiális proteinek szélesebb körű jelenlétéről a fonalasgombák körében. A szerkezet-hatásfok összefüggés feltárása, mind elméleti, mind gyakorlati szempontból fontos lehet, lehetőséget teremtve új, hatékony antimikrobiális szerek kifejlesztésére. Kitűzött céljaink teljesítése mikrobiológiai problémákon túlmutató kérdésekre is választ adhat.

The defensins and similar antimicrobial proteins are widely distributed in the nature. Five similar proteins have been identified among filamentous fungi. These proteins secreted by taxonomical distinct species have different mode of action and species specificity, nevertheless their structure is very similar. Neosartorya fischeri antimicrobial protein (NFAP) from N. fischeri (anamorf: Aspergillus fischerianus) is a hypothetical antifungal protein derived from genomic database, its presence is confirmed by in silico investigation only. Its cloning, isolating, antifungal spectrum, mode of action and biological role has not been described yet. The goal of the proposed project is to increase the knowledge of defensin-like antimicrobial proteins derived from filamentous fungi by investigation of NFAP.
The main aims of the research project are the followings: (1) determination of the presence of NFAP and its orthologs from Neosartorya fischeri (NRRL 181) and related species; (2) investigation of the antimicrobial properties and mode of the action in case of NFAP; (3) creation a heterologous expression system for production of NFAP; (4) examination of the connection between the structure of NAFP and its efficacy; (5) observation of biological role of NFAP.
The final results provide information about the distribution of defensin-like antimicrobial proteins among filamentous fungi. The results about the structure-efficacy connection will be the bases of further theoretical and practical studies to develop effective antimicrobial drugs. Achievements of this project will provide important information in addition to microbiology.

A projekt célja a Neosartorya fonalasgomba nemzetség által termelt defenzin-szerű antimikrobiális proteinek azonosítása és jellemzése volt a Neosarorya fischeri NRRL 181 antifungális protein (NFAP) átfogó vizsgálatán keresztül. A projekt során NFAP homológ proteineket kódoló géneket azonosítottunk öt Neosartorya izolátum esetén. Meghatároztuk az NFAP antimikrobiális spektrumát és antifungális tulajdonságait eltérő környezeti körülmények között. Megvalósítottuk az NFAP heterológ expresszióját Pichia pastoris-ban és Aspergillus nidulans-ban, előbbivel az NFAP nagy mennyiségben termeltethetővé vált. Bizonyítottuk, hogy az NFAP központi hidrofób magjának szerepe lehet az antifungálisan aktív szerkezet kialakításában. Kimutattuk, hogy az NFAP képes elektrosztatikusan kötődni az érzékeny gombához, annak membránját szétrombolni és reaktív oxigéngyökök indukcióján keresztül apoptózist és nekrózist kiváltani. Bizonyítottuk, hogy az NFAP jelenlétében a szenzitív gomba metabolikusan inaktívvá válik és zavart szenved a sejtfal kiépülése. Vizsgálataink alapján az NFAP hatásmechanizmusa hasonlóságot mutat az Aspergillus giganteus AFP és a Penicillium chrysogenum PAF esetén leírtakkal, de azoktól eltérően egy ismeretlen célpontja is lehet. Megfigyeltük, hogy az NFAP szerepet játszik az tápanyagokért és az élőhelyért folytatott versenyben a hasonló ökológiai niche-t elfoglaló gombafajokkal szemben. Eredményeink hozzájárulhatnak peptid-alapú antifungális szerek kifejlesztéséhez.

The aim of the present project was the identification and characterization of defensin-like antimicrobial proteins secreted by Neosartorya filamentous fungal genus by comprehensive investigation of Neosarorya fischeri NRRL 181 antifungal protein (NFAP). In this project NFAP homolog protein encoding genes were identified in five Neosartorya isolates. The antifungal spectrum and antifungal features of NFAP within different environmental conditions was determined. The heterologous expression of NFAP was carried out in Pichia pastoris and Aspergillus nidulans. High yield of NFAP was achieved by using P. pastoris. Our results revealed that the central hydrophobic core of NFAP may play a role in the proper folding of the antifungally active structure. It was observed that NFAP can bind electrostatically to the sensitive fungus and destroys its cell membrane, furthermore triggers apoptotic-necrotic event through reactive oxygen species. In the presence of NFAP the sensitive fungus suffered metabolic inactivity and the organization of the cell wall was damaged. Based on our previous studies it was concluded that the antifungal mechanism of NFAP shows similarities to that of Aspergillus giganteus AFP and Penicillium chrysogenum PAF, but an unknown cellular target is hypothesised. It was demonstrated that NFAP plays a role in the emulation for nutrients and habitat against fungi with similar ecological niche. Our results contribute to development of peptide-based antifungal agents.
