A növényi hormonoknak a betegségek vagy a rezisztencia elősegítésében játszott szerepét a legutolsó időkig viszonylag elhanyagolták a kutatások. Másrészről, egyéb növekedés-szabályozó anyagokat, mint a szalicilsav (SA), jázmonsav (JA) és a brasszinoszteroidok, fontos betegség- és stressz-rezisztencia faktoroknak tekintenek.
Ismert, hogy a reaktív oxigén fajták (ROS) fontos szerepet játszanak a betegség-ellenállóságban, és a növényi hormonok erősen befolyásolják a ROS képződésétés az antioxidánsokat. A SA és JA a növényi védekezés jelátvivő molekulái, és a SA által közvetített rezisztencia a biotróf, míg a JA által közvetített rezisztencia inkább a nekrotróf kórokozók és a kártevő rovarok ellen hatékony.
Fontos kérdés, hogy a különböző hormonok hogyan hatnak egymás jelátviteli rendszerére a betegség és a rezisztencia kialakulása során? Nevezetesen, hogyan befolyásolják a citokininek és a brasszinoszteroidok a JA/etilén vagy a SA által közvetített növényi védekezési reakciót? A projekt célja a fenti növényi hormonoknak a biotróf és nekrotróf kórokozók elleni rezisztenciára gyakorolt szinergikus és antagonisztikus hatásának jellemzése hormon túltermelő és hormon hiányos transzgenikus és mutáns dohány és Arabidopsis thaliana vonalak segítségével. A SA és a JA/etilén jelátviteli utak PR fehérje génjeinek indukcióját, vagy gátlását, illetve a ROS és antioxidánsok változását is megvizsgáljuk. Továbbá „Shooter” és „Rooter” Agrobacterium tumefaciens mutánsok, valamint agroinfiltrációs módszer alkalmazásával a citokinin és auxin bioszintézis tranziens növelésének a biotróf és nekrotróf kórokozókkal szembeni rezisztenciára gyakorolt hatását tanulmányozzuk.
Summary
The role of plant hormones in pathogenesis and resistance was relatively neglected until recently. On the other hand, other plant growth regulating substances such as salicylic acid (SA), jasmonats (JA) and brassinosteroids (BR) have been considered as important factors of disease and stress resistance of plants.
It is known that reactive oxygen species (ROS) play an important role in disease resistance of plants, and plant hormones can strongly influence induction of ROS and antioxidants. SA and JA are signalling molecules in plant defence and SA-mediated resistance is effective against biotrophs, whereas JA- or ethylene-mediated responses are predominantly active against necrotrophs and herbivorous insects.
It is an important question that how the various hormone signalling interacts during pathogenesis or resistance. Namely, how can cytokinins and brassinosteroids influence JA/ethylene and SA mediated responses of plants to necrotrophic and biotrophic pathogens? The aim of this project is to characterize the synergistic or antagonistic effects of the above plant hormones on resistance of plants by applying hormone overproducing or deficient transgenic or mutant tobacco and Arabidopsis thaliana lines. The up or down regulation of genes of PR proteins of SA and JA/ethylene pathways will also be checked, in addition to the changes in ROS and antioxidants. In addition, „Shooter” and „Rooter” Agrobacterium tumefaciens mutants will be applied to tobacco leaves by agroinfiltration method in order to analyze the effect of the transient expression of cytokinin and auxin biosynthesis genes on necrotrophic and biotrophic interactions.

Munkánk célja az volt, hogy jobban megismerjük a növényi hormonok szerepét a különböző gazdanövény-kórokozó kapcsolatokban. Kimutattuk, hogy a brasszinoszteroid (BR) és a progeszteron előkezelés jelentősen csökkentette a P. syringae pv. syringae baktérium indukálta ion kiáramlást, a II. fotoszisztéma és hőkibocsátás változásokat repce, illetve Arabidopsis thaliana növényeken. Kimutattuk, hogy a fény fokozza az etilén és az etán termelődését, különösen roncsolt növényi szövetekben. Bebizonyítottuk, hogy a Piriformospora indica, mikorrhiza-szerű gomba szignifikánsan csökkentette a Fusarium culmorum kórokozó által okozott károsítást árpában, szoros összefüggésben a kórokozó mennyiségének csökkenésével. A fuzárium fertőzés hatására megnőtt a gyökerekben a lipidperoxidáció és csökkent az enzimatikus és nem enzimatikus antioxidáns aktivitás, amely hatásokat kivédte az előfertőzés P. indica gombával. Inkompatibilis és kompatibilis paprika-tobamovirus gazda-parazita kapcsolatokban végzetünk hormon és cukortartalom meghatározásokat, kiegészítve brasszinoszteroid hormon kezeléssel. Megállapítottuk, hogy nemcsak a tipikus stressz hormonok (SA, JA, ABA), hanem a citokininek, auxinok és gibberelinek mennyisége is szignifikánsan változott. Hasonlóan, a szénhidrát analízis is jelentős változásokat mutatott vírus fertőzés hatására.
Results in English
The aim of the project was to get a deeper insight into the role of plant hormones in various plant-pathogen interactions. We demonstrated that brassinosteroid (BR) and progesterone pre-treatments remarkably decreased ion leakage, and changes of photosystem II efficiency as well as of heat flow in rape or in Arabidopsis thaliana. It was also shown that light increased ethylene and ethane production especially in damaged tissues. We proved Piriformospora indica, a mycorrhiza-like fungus significantly decreased the damage caused by the pathogen Fusarium culmorum on barley, in connection with the decreased relative amount of fungal DNA. Fusarium infection elevated the rate of lipidperoxidation, decreased the enzymatic and non-enzymatic antioxidant activities in the root, but these effects were alleviated by P. indica pre-treatment. We wrote a review article on the role of reactive oxygen species (ROS) and plant hormones in plant resistance against necrotrophic and biotrophic pathogens. Sugars, as well as hormone contents were studied in pepper leaves infected with a compatible or an incompatible tobamovirus by UHPLC-MS, or HPLC methods, supplemented with BR pre-treatment. Importantly, not only the amount of typical stress hormones like SA, JA or ABA changed significantly, but also the amount of cytokinins, auxins and gibberellins. Similarly, the sugar analysis showed remarkable changes after virus infections.
