Az invazív candidiasis korunk egyik növekvő, a Föld egész népességét érintő egészségügyi problémája. A Candida albicans után a Candida parapsilosis a leggyakoribb kiváltója a betegségnek, és bár a két faj mind genetikájában, virulencia faktoraiban, epidemiológiájában, az általuk előidézett immunválaszban és hatóanyagérzékenységében eltér egymástól, a C. parapsilosis kutatások mindezidáig a háttérbe szorultak. Elsőként vezettük be a C. parapsilosis kutatásban az irányított géndeléciós eljárást, valamint létrehoztunk és validáltunk rekonstruált emberi szövettenyészetet alkalmazó fertőzési teszt modelleket a patogenitás vizsgálata céljából. Eddigi sikeres kutatásaink egyértelműen alátámasztják a kutatási területet érintő kompetenciánkat és biztosítják a jelen pályázat sikeres végrehajtását. A pályázatunkban a C. parapsilosis patogenezisében szerepet játszó molekuláris mechanizmusokat kívánjuk feltárni géndeléciós mutáns törzsek létrehozásával, ezek virulenciájának tanulmányozásával emberi rekonstruáltszövetek, makrofág sejtvonalak in vitro fertőzési modelljein keresztül, a mutáns és vad típusú törzsek fertőzéskor végbemenő génexpressziós eltéréseinek tanulmányozásával, a humán monociták/makrofágok transzkriptóm változásának vizsgálatával vad és mutáns C. parapsilosis fertőzések során, és végül a szekretált enzimek szerkezetének tanulmányozásával potenciális gátlószerek azonosítása céljából. Ezen vizsgálatok OTKA támogatása egy hatékony kutatócsoport erős megalapozását jelentené a C. parapsilosis kutatás élvonalában, valamint az eredmények a C. parapsilosis patobiológiájának teljes és komplex megértéséhez vezetnek el, mely lehetőséget nyithat újszerű, C. parapsilosis ellenes specifikus hatóanyagok felfedezésére terápiás alkalmazás céljából. Mindazonáltal a projekt a C. parapsilosis egyedi biológiai tulajdonságainak - amelyek alkalmassá teszik a modern klinikai környezethez való alkalmazkodásra - megismerése révén, az alapkutatás számára is hasznos új ismereteket szolgáltathat.

Invasive candidiasis is a major global health problem and Candida parapsilosis is currently the most common cause of invasive candidiasis worldwide after C. albicans. Importantly, there are significant basic differences in genetics, epidemiology, virulence determinants, and drug susceptibilities between C. parapsilosis and C. albicans. Although C. albicans is a broadly studied pathogen, the pathobiology of C. parapsilosis is poorly understood, particularly in regard to the genetic basis of the virulence. Notably, I provided a major breakthrough in the study of C. parapsilosis pathogenesis by developing the first targeted gene deletion technique for this organism and I also established and validated several pathogenicity testing systems, including reconstituted human tissue and murine models. These successes clearly reflect my competence in the field and ensure the successful execution of the proposed OTKA NF project, which focuses on the role of secreted hydrolytic enzymes in the pathogenesis of C. parapsilosis by generating gene disruptants; virulence in human tissue and in vivo infection models; differential gene expression of the generated mutants; whole transcriptome dynamics of human monocytes/macrophages upon C. parapsilosis wild type and mutant infection; protein structures and potential inhibitory compounds of the secreted fungal enzymes. These studies will definitely lead to the rapid growth in our knowledge on every aspect of C. parapsilosis pathobiology. Funding of this application by OTKA will solidly establish a Hungarian laboratory at the forefront of the C. parapsilosis field and the fundamental knowledge that we will discover on the biology of C. parapsilosis will lead to the identification and analysis of molecules and genes involved in virulence and pathogenicity of this important fungal pathogen.

A projekt keretében elvégzett kutatások eredményeként jelentősen bővültek ismereteink az opportunista patogén gombák virulenciájának és patomechanizmusainak vizsgálatának területén. In vitro és in vivo modellrendszereket dolgoztunk ki a Candida parapsilosis virulenciájának vizsgálatára. Megállapítottuk a gomba szekretált lipázainak szerepét a gazda immunválaszának modulálásában. Elsőként bizonyítottuk, hogy a Candida parapsilosi által kiváltott T sejtes immunválasz jelentősen eltér a Candida albicans-étól. Elemeztük a makrofágok fagocitózis folyamatait in vitro Candida parapsilosis fertőzések során. Bizonyítottuk a defenzin szerű molekulák szerepét a Candida ellenes gazdaválaszban. Továbbá összehasonlító genomikai vizsgálatokkal igazoltuk, hogy a Candida parapsilosis fajkomplex tagjai képesek hibridek létrehozására, amely folyamatnak a patogénné válás során is szerepe lehet.

Candida albicans (Ca) and Candida parapsilosis (Cp) are the two main yeast species that are both commensal of humans and responsible for devastating disseminated infections. The aim of this NF84006 project was to investigate the virulence and the pathomechanisms of the important human fungal pathogen Candida parapsilosis. First we established and further developed in vitro and in vivo infection models to examine host pathogen interactions. We adapted in vitro models using cell lines (J774.2 murine macrophages, THP-1 human macrophages) or primary cells (Bone marrow derived macrophages from mouse, peripherial mononuclear cell derived macrophages from human). We showed that fungal lipases can reduce host inflammatory response and proved their role in the pathogenesis. We investigated the cellular immune response orchestred by phagocytes. We proved the crucial role of defensins in the host anti-Candida response and we were the first to show the major differences in the T cell response upon Candida parapsilosis infections in comparison to Candida albicans in vitro. We also investigated the recent genomic evolution within the C. parapsilosis sensu lato complex, and identified hybrids in the species complex.
