A legutóbbi OTKA pályázatunk (K49276) során felfedeztük, hogy a Thiocapsa roseopersicina hidrogenáza autokatalitikus reakcióban bontja a hidrogén molekulát, mutatva az autokatalitikus reakciók alapvető tulajdonságait; spontán képződő, állandó sebességgel terjedő reakciófrontok alakulnak ki vékony oldatrétegben. Megállapítottuk, hogy az enzimreakció egyedülálló abban az értelemben, hogy az autokatalizátor az enzim egyik konformere. A reakciókörülmények megfelelő módosításával sikerült időben egyre csökkenő frekvenciájú oszcillációt is kimutatnunk.
A pályázat célja a hidrogenáz autokatalitikus reakciójának szisztematikus megismerése. Ez magába foglalja a zárt rendszerben tapasztalt oszcilláció és, megfelelő reaktor tervezése után, a nyílt rendszerben megvalósíthatónak tűnő állandósult oszcilláció tanulmányozását. Ezzel párhuzamosan egy, a kísérleti eredményekkel összhangban levő modellt tervezünk felállítani, melynek nyílt rendszerben történő vizsgálatával lehetőség nyílik az egyes fizikai és kémiai paraméterek hatásának előrejelzésére.
A betáplált hidrogéngáz és a benzil-viologén oxidált formájának különböző kiindulási mennyisége mellett a reakció pH függését felhasználva, a kezdeti pH-t és a reakciósebességet befolyásoló hőmérsékletet változtatva feltérképezzük a rendszer tér- és időbeli viselkedését.
Célunk annak megértése, hogy egy enzim miként szerepelhet autokatalizátorként és ennek milyen biokémiai következményei lehetnek.
Summary
Supported by our last, OTKA financed grant (K49276) we discovered that the HynSL hydrogenase from Thiocapsa roseopersicina splits molecular hydrogen in an autocatalytic reaction, as evidenced by the typical characteristics of such reactions: in an unstirred solution spontaneously formed reaction fronts propagate at a constant velocity. The reaction is unique in the sense that the autocatalyst is a conformer of the hydrogenase enzyme.
The aim of this grant proposal is to continue the systematic characterization of the autocatalytic reaction of the hydrogenase enzyme in order to obtain the mechanism and the full phase diagram of the enzyme reaction cycle.
The investigation of the observed oscillations in a closed system and, after designing a proper open reaction vessel, studies of the possible permanent oscillations therein are also planned. Parallel theoretical calculations and model construction will enable us to predict the effect of basic physical and chemical parameters on the oscillatory behavior both in open and closed systems.
The effect of pH, oxidized benzyl viologen and hydrogen gas concentrations on the reaction as well as the temperature dependence of the reaction kinetics will be investigated in order to describe the spatiotemporal behavior of the system.
Our aim is to elucidate the autocatalytic role of the enzyme and the biochemical consequences it may hold.

Technikai fejlesztések 1. Kifejlesztettünk egy új reakciókamrát (TC-TLRC). 2. Az új reakcióedényben lehetővé vált ugyanannak a hidrogenáz mintának a használata mindkét reakcióirányban. Tudományos eredmények 1. Megmagyaráztuk a hidrogenáz aktiválás folyamatát. 2. Megmutattuk, hogy a hagyományos hidrogenáz model önellentmondásokat tartalmaz. 3. Kimutattuk, hogy a hidrogenáz katalitikus ciklusában egy autokatalitikus reakciónak kell lennie. 4. Oszcilláló reakciót mutattunk ki a hidrogenáz katalitikus ciklusában. 5. Három különböző hőmérsékleti tartományt azonosítottunk, ahol a hidrogenáz reakció különbözőképpen zajlik: ~8-14 ºC, 14-28 ºC és 30 ºC felett. 6. A H2 koncentráció növekedésével az első és a második ciklusban mért aktivitások és látszólagos szaturációs értékek különbsége növekedett. 7. Kozmotróp és semleges Hofmeister sók a hidrogenáz aktivitását nem változtatták. A kaotróp sók a H2 oxidáló aktivitásra nem voltak hatással, viszont a proton redukáló hatást teljesen leblokkolták. 8. A benzil viologén elektrokémiai viselkedése drasztikusan megváltozott kaotróp sók jelenlétében. 9. Meghatároztuk a metil és benzil viologének kötőhelyeit a hidrogenáz felületén. 10. A D. gigas esetében a autokatalitikus reakció mindazon jellegzetességeit megtaláltuk, amelyeket a Thiocapsa roseopersicina hidrogenáza esetében megfigyeltünk. A másik hidrogenáz a Ralstonia eutropha oxigéntűrő hidrogenáza volt. Itt az enzimkoncentráció függő lag fázis jelenlétét tudtuk kimutatni

Technical achievements 1. A new reaction chamber (TC-TLRC) was designed and built. 2. It became possible to run the reaction back and forth several times. Scientific achievements 1. We explained the activation of the hydrogenase as an autocatalytic reaction. 2. The traditional hydrogenase models are contradictory and cannot explain the activation under hydrogen on hydrogenase. 3. We have shown that there is an autocatalytic step in the enzyme cycle of hydrogenase 4. Oscillations can be initiated in the hydrogenase reaction. 5. Three different temperature regions were identified depending on the kinetic and light scattering behavior of the enzyme: ~8-14 ºC, 14-28 ºC and above 30 ºC. 6. When the H2 substrate concentration increased, the activity and saturation level difference between the first cycle and the other two cycles also increased 7. Kozmotropic and neutral salts had no effect on the activity of hydrogenase, whereas chaotropic salts blocked proton reduction. 8. Benzyl viologen dramatically changed its electrochemical properties when chaotropic salt was present. 9. The binding sites of the benzyl- and methyl viologen molecules to the surface of the hydrogenases were calculated. 10. All the characteristic properties of an autocatalytic reaction found in T. roseopersicina hydrogenase were also present in the case of D.gigas hydrogenase. The oxygen tolerant hydrogenase of R. eutropha showed enzyme concentration dependent lag phase.
