A sejtek genetikai állományának őrzését speciális fehérjék együttese végzi, amelyek asszociátumokat képezve a nukleinsavakkal felismerik és kijavítják a genetikai hibákat. Rákos sejtek terápiája esetén viszont ezek a javító mechanizmusok a kívánt sejtpusztítás ellen dolgoznak. A projekt célja az alapvető DNS-hiba javító mechanizmus, a homológ rekombináció lépéseiben szerepet játszó nukleinsav-szerkezeteknek és nukleoprotein komplexeknek a centrális szerepet betöltő rekombináz, a humán Rad51 általi felismerését szabályozó kölcsönhatások szerkezeti és energetikai jellemzése. Az alkalmazandó modern és unikális biofizikai kísérleti technikák alkalmasak a speciális szerkezetekhez kötött fehérjék kötőhelyének mechanikai, szerkezeti, kinetikai és dinamikai tulajdonságainak jellemzésére és az egyes kötődési reakciók termodinamikai paramétereinek meghatározására. A vizsgálatokat a hidrosztatikai nyomás és a hőmérséklet függvényében végzett fluoreszcencia spektroszkópia, izotermikus titrációs kalorimetria, gyors kinetikai valamint egy-molekula érzékenységű fluoreszcencia mikroszkópiai, atomi erő mikroszkópiai és lézer-csipesz módszerek segítségével végezzük. A munkacsoport számára rendelkezésre álló modern módszerek egyedülállóan széles skálája lehetőséget ad az eredeti kísérleti megközelítésekre, és a résztvevő minősített kutatók gyakorlata garancia az új, izgalmas eredmények elérésére.
Summary
The maintainance and stability of the genetic material of the cells is of vital significance. This task is fulfilled by the associates of specific proteins and DNA complexes that recognize and repair the DNA damages. In cancer cells, however, the very same mechanisms oppose the effect of chemo- or radation- therapy since these are aimed to destroy the genetic material. In this project we plan to study those structural conditions that are detrimental in one of the basic repair processes, homologous recombination in building up the functional nucleoprotein associates of human Rad51. This protein plays key role as a recombinase in human cells. We propose to implement unique experimental approaches of modern fluorescence spectroscopy, microscopy, calorimetry and single molecule studies that became recently available in the Department. These methods represent a unique combination of approaches that has not been applied in this field, and the participants are experts of these techniques. We suggest to structurally characterize the binding sites of the protein for self association, for association with regulating other proteins, primarily with HLTF, and with DNA segments, and determine the mechanical, dynamic, kinetic and crowding conditions of the macromolecular complex formation processes of vital significance.

A projekt Kidolgozása során új biokémiai laboratóriumot hoztunk létre a homológ rekombinációban alapvető szerepet játszó humán Rad51 rekombináz előállítására és a fizikai mérések előkészítésére. A molekuláris klónozástól elindulva újraterveztünk és megvalósítottunk egy fehérje termeltetési és tisztítási eljárást, amellyel a hRad51 fehérje vad tipusát és négyféle triptofán-mutánsát spektroszkópiai tisztaságban és kb 50 M koncentrációban elő lehet állítani. Új mérőberendezést állítottunk össze és szoftvert fejlesztettünk az adatok kiértékeléséhez, amelynek segítségével a rekombináz funkcionális filamentumait felépítő monomérek határfelületi kötéserőssége meghatározható. Kimutattuk, hogy a DNS-hibajavításban kulcsfontosságú preszinaptikus filamentum szerkezete csak ATP-t kötve képes ellátni a funkcióját, és kimutattuk ennek a szerkezetnek a kiemelkedően erős stabilitását. Az általunk kidolgozott termeltetési és tisztítási eljárással előállított fehérje, a hRad51 alkalmas további fehérje önasszociációs és fehérje-DNS kötődési jelenségek tanulmányozására. Sikeres előkísérletek alapján elindítottunk két újabb kutatási irányt : A hRad51 triptofán mutánsai alkalmasak a funkcionális filamentum felépülési kinetikájának vizsgálatára; a dsDNS-re ép ülő filamentum szerkezete AFM-mérésekkel vizsgálható.
Results in English
During the elaboration of the project, a new laboratory was established and equipped for the production and purification of the wt human recombinase, Rad51. We tackled the problems arising from the extremely strong tendency of this protein for association. We needed to implement and modify the described protocols starting from cloning and to reach spectroscopic purity and high concentration as ~ 50 M. The procedure was elaborated for the wt protein and for four triptophan mutants. We constructed a new device and developed appropriate evaluation software by which the interface binding strength of protein oligomers can be determined. We applied this method for the functional presynaptic filament (PSF) of wt hRad51 built on ssDNA and for its other oligomer states. We determined the interface binding strength in various oligomer states and showed the detrimental role of ATP-binding in the functional structural form and in the stability of PSF. By having hRad51 samples available, we established the possibility for studying the conditions of protein self-association and DNA-binding reactions of outmost significance. We started two new lines of studies based on successful preliminary experiments. The kinetics of oligomerization can be studied by the signal of Trp-mutants (M.Kovacs, ELTE TTK), oligomerization on dsDNA could be successfully studied by using the AFM techniques available in the Institute (M.Z. Kellermayer).
